S-美芬妥英羟化酶在云南白族人群中的遗传多态性

摘要 研究云南白族人群中S-美芬妥英羟化酶活性的遗传多态性。随机选择互无亲缘关系的健康白族志愿者口服美芬妥英100mg,收集服药后0~8h尿,分别用反相高效液相色谱紫外检测方法和手性毛细管气相色谱氮磷检测方法定量分析其尿中氧化代谢产物4′-羟基美芬妥英和S/R-美芬妥英比值。结果:在该少数民族人群中,S-美芬妥英羟化酶活性的人群分布呈遗传多态性。 To investigate the genetic polymorphism of S-mephenytoin 4′-hydroxylase (CYP2C19) in a Chinese Bai population,204 unrelated healthy Bai Chinese including both sexes were randomly selected and studied in this paper.Each of them was given 100 mg of mephenyltoin tablet after emptying the bladder and their urine samples were collected within 8 hours of the postdose.S-and R-mephenyltoins in urine were detected by chirocapillary gas chromatography with nitrogen-phosphorous method,and 4′-hydroxymephenytoin,the oxidative metabolite,by reversed phase liquid chromatography with ultraviolet method.The activity of enzyme CYP2C19 was expressed as S/R ratio.The S/R ratio above an antimode of 0.70 was phenotyped to be a PM.The results show that the polymorphism of CYP2C19 activity exist in Bai population,a minority in china.15% of the population show a genetic deficiency in the activity of PMs.
