葡萄籽原花青素对糖尿病大鼠肾脏GLUT2及肾功能的影响 被引量:6

摘要 目的:观察葡萄籽原花青素对糖尿病大鼠肾组织GLUT2及肾功能的影响。方法:将SD雄性大鼠随机分为对照组、糖尿病模型组、两个葡萄籽原花青素组。两个葡萄糖花青素组分别给予200mg·kg^(-1)·d^(-1)、400mg·kg^(-1)·d^(-1)灌胃,对照组和模型组灌等体积的生理盐水,8周后测定各组大鼠血NO、Ccr、尿蛋白定量,并处死大鼠取肾检测肾脏肥大指数,以免疫组化和免疫荧光技术检测肾组织GLUT2的表达。采用酶联免疫吸附法检测血清白细胞介素8、转化生长因子。结果:与对照组比较,糖尿病模型组大鼠肾脏GLUT2表达明显增加,NO水平、Ccr、尿蛋白定量、IL-8、TGF-β、肾脏肥大指数显著升高(P<0.05),NO/ET显著降低(P<0.05);与模型组比较,葡萄籽原花青素组大鼠肾脏GLUT2表达明显减少,NO水平、Ccr、尿蛋白定量、IL-8、TGF-β、肾脏肥大指数活性显著降低(P<0.05),NO/ET显著增高(P<0.05)。结论:葡萄籽原花青素能够降低肾脏GLUT2表达,在一定程度上改善DN模型大鼠肾脏损伤。 Objective To observe the effect of grape seed procyanidins on renal tissue GLUT2 and renal function in diabetic rats.Methods: SD male rats were randomly divided into the control group, the diabetes model group, and two grape seed procyanidins groups(the low dose group received 200mg·kg-1·d-1, the high dose group received 400mg·kg-1·d-1). Two grape seed procyanidins groupswere respectively treated with corresponding dosage. The control group and the diabetes model group were given equal volume of saline.In 8 weeks, the blood NO, Ccr and urine protein quantitative were measured in each group. After rats were killed, kidney hypertrophyindex was detected; and renal tissue GLUT2 was detected by immunofluorescence technique. IL-8, TGF were detected by enzyme linkedimmunosorbent assay. Results: Compared with the control group, renal tissue GLUT2 expression was significantly increased in two grapeseed procyanidins groups, levels of NO, Ccr, urine protein quantitative, IL-8, TGF-β and kidney hypertrophy index were significantlyincreased (P〈0.05). NO/ET was significantly lower (P〈0.05). Conclusion: Grape seed procyanidins can reduce the expression of renaltissue GLUT2 and improve the renal damage of DN model partly.
