摘要
目的探讨远志皂苷(TEN)对转运蛋白RAGE和LRP1的调控作用,以及对Aβ诱导的PC12细胞氧化应激的保护作用。方法建立Aβ_(25-35)诱导的PC12细胞损伤模型,MTT法测定细胞活力,Western blot检测RAGE和LRP1蛋白表达水平变化,分光光度法检测活性氧(ROS)、丙二醛(MDA)浓度及超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性变化。结果 TEN能明显下调RAGE的表达,上调LRP1的表达,并且抑制Aβ_(25-35)诱导引起的ROS、MDA浓度增加,提升Aβ_(25-35)诱导引起的SOD、GSH-Px活性降低。结论 TEN可能通过调控Aβ转运清除蛋白RAGE和LRP1的表达,抑制Aβ诱导的氧化应激效应,发挥抗Aβ神经毒性和神经保护作用。
Objective To investigate the effect of tenuigenin( TEN) in regulation of RAGE and LRP1 expression,and its protective effect for Aβ_(25-35)-induced oxidative stress in PC12 cells. Methods The PC12 cell injury model was established by Aβ_(25-35) stimulation. The cell viability was analyzed by MTT. The expression of RAGE and LRP1 was detected by western blot. The concentration of ROS and MDA and the activity of SOD and GSH-Px were detected by spectrophotometry. Results Treatment with TEN significantly reduced RAGE expression,but increased the LRP1 expression. TEN inhibited the levels of ROS and MDA,but increased the activities of SOD and GSH-Px in Aβ_(25-35)-stimulated PC12 cells. Conclusion TEN can inhibit Aβ_(25-35)-induced oxidative stress in PC12 cells by different regulation of RAGE and LRP1 expression.
