佛手提取物调控CYP7A1蛋白表达的研究 被引量:9

摘要 目的探讨佛手提取物(Finger Citron extract)对Hep G2细胞中胆固醇7α羟化酶(CYP7A1)及其核转录调控因子过氧化物酶体增殖物激活受体α(PPARα)和肝核因子4α(HNF4α)的蛋白表达量的影响。方法采用MTT法得出佛手提取物对Hep G2细胞生长无明显抑制的浓度,Western blot检测CYP7A1、PPARα和HNF4α的蛋白表达量;采用siRNA干扰技术沉默调控因子PPARα后,观察佛手提取物对CYP7A1的蛋白表达的影响。结果佛手提取物浓度为0.2、1、5、25μg/m L对Hep G2细胞无明显的抑制作用,且均能上调CYP7A1、PPARα和HNF4α的蛋白表达量,沉默调控因子PPARα后,CYP7A1的蛋白表达量显著下降。结论佛手提取物具有一定的降脂作用,其机制可能与其上调PPARα的表达来上调CYP7A1的表达有关。 Objective To investigate the effect of Finger Citron extract( FCE) on the expression of cholesterol 7 alpha hydroxylase( CYP7A1),nuclear regulation factor peroxisome proliferator activated receptor alpha( PPARα) and hepatocyte nuclear factor 4 alpha( HNF4α) on Hep G2 cells. Methods MTT assay was used to evaluate the effect of FCE on the viability of Hep G2 cells. The expression of CYP7A1,PPARα and HNF4α was tested by western blot. siRNA interference technology was used to silence the transcription factor of PPARα,and then the effect of FCE on CYP7A1 protein expression was examined.Results FCE at 0. 2,1,5,25 μg / m L did not show obviously inhibitory effect on the viability of Hep G2 cells,but increased the expression of CYP7A1,PPARα and HNF4α. After silencing the transcription factor PPARα,the expression of CYP7A1 was decreased. Conclusion The lipid-lowering effect of FCE may be related to upregulation of CYP7A1 via PPARα pathway.
