摘要
目的:观察小檗碱和吴茱萸碱配伍对高脂血症模型大鼠肝脏HMG-CoA还原酶和L-FABP基因表达的影响,探讨黄连吴茱萸配伍发挥降脂作用的机制。方法:将40只健康雄性SD大鼠随机分为正常对照组,模型对照组,辛伐他汀组,小檗碱与吴茱萸碱配伍高、低剂量组,每组8只;使用高脂乳剂建立大鼠高脂血症模型,以18sRNA作为内参基因,RT-PCR技术检测各组大鼠肝组织中HMG-CoA还原酶和L-FABP mRNA相对表达水平。结果:小檗碱和吴茱萸碱配伍能显著降低高脂血症模型大鼠肝脏HMG-CoA还原酶和L-FABP mRNA表达,且高剂量组效果更优。结论:黄连吴茱萸降血脂有效成分小檗碱、吴茱萸碱配伍可降低高脂血症模型大鼠的血脂水平,其作用机制可能与抑制肝脏HMG-CoA还原酶和L-FABP基因表达有关。
Objective: To observe the effects of berberine combined with evodiamine on liver HMG-CoA reductase and L- FABP mRNA expression levels in a rat model of hyperlipidemia and to investigate the lipid- lowering mechanism. Methods: A total of 40 healthy male Sprague- Dawley rats were equally and randomly divided into normal control group,model control group,simvastatin group,and low- and high- dose berberine combined with evodiamine groups. The rat model of hyperlipidemia was established using high- fat emulsion. The relative expression levels of liver HMG- CoA reductase and L- FABP mRNA were measured using RT- PCR with 18 sRNA as a reference gene in these different groups. Results: Berberine combined with evodiamine significantly reduced the HMG- CoA reductase and L- FABP mRNA levels,and the high- dose group had greater reductions in the two indices. Conclusion:Berberine combined with evodiamine can reduce blood lipid level in the rat model of hyperlipidemia and its mechanism may be related to the inhibition of liver HMG- CoA reductase and L- FABP mRNA expression levels.
