摘要
目的探讨炭疽毒素受体1(anthrax toxin receptor 1,ANTXR1)蛋白在食管鳞状细胞癌(esophageal squamous cell carcinoma,ESCC)组织中的表达情况及意义。方法应用实时荧光定量聚合酶链反应(real-time polymerase chain reaction,real-time PCR)技术和免疫组织化学法检测ESCC和相应癌旁正常组织中ANTXR1蛋白的表达水平,分析其表达与患者临床病理各参数的关系。结果在10对新鲜食管癌组织中,mRNA水平检测ANTXR1在ESCC组织中的表达明显高于癌旁正常组织(P<0.01);在172例石蜡标本中,ANTXR1蛋白在ESCC组织的阳性表达率明显高于癌旁正常组织,且ANTXR1蛋白的表达与ESCC组织分化程度、有无淋巴结转移、肿瘤分期以及患者的预后相关(P<0.05),与患者的年龄、性别、肿瘤大小和肿瘤的浸润深度无相关性(P>0.05)。结论 ANTXR1蛋白在ESCC组织中高表达,并且与患者的预后相关;ANTXR1蛋白可能是新的ESCC促癌因子,参与ESCC发生、发展,并有望为ESCC的预后判断和治疗提供新的思路。
Objective To investigate the expression and significance of ANTXR1 protein in esophageal squamous cell carcinoma(ESCC)tissues.Methods The expression of ANTXR1 mRNA and protein in ESCC tissues and adjacent normal tissues were detected by real-time polymerase chain reaction(real-time PCR)and immunohistochemistry.Results In 10 pairs of the fresh ESCC tissues,the mRNA level of expression of ANTXR1 was higher than that of the adjacent normal tissues(P〈0.01).The positive rate of expression of ANTXR1 protein in 172 cases of the ESCC sections was significantly higher than that in the adjacent normal sections(P〈0.01).The expression of ANTXR1 protein was significantly correlated with histological differentiation,lymph node metastasis,neoplasm staging and clinic prognosis(P〈0.05),but not with age,gender,infiltration degree and size of tumor(P〉0.05).Conclusion The high expression of ANTXR1 protein in ESCC tissues and ANTXR1 expression is related to the patients' prognosis.ANTXR1 may be a novel oncogene relating to the tumorigenesis and tumor progression of ESCC,and could supply new ideas for the prognosis and treatment of ESCC.
