摘要 目的:运用中药血清药理学方法研究柴胡疏肝散含药血清对内质网应激的保护作用并探讨其机制。方法:实验用衣霉素(TM)对NG108-15神经细胞进行诱导,建立内质网应激(ERS)细胞模型,柴胡疏肝散含药血清为干预组,采用逆转录聚合酶链式反应方法观察泛素(Ub)的m RNA表达水平。结果:各浓度柴胡疏肝散含药血清干预组Ub m RNA表达与模型组相比,表达水平明显上调,差异具有统计学意义(P<0.05)。结论:柴胡疏肝散方药对内质网应激引起的神经元损伤有显著的保护作用,其机制可能与泛素-蛋白酶体通路(UPP)相关联。 Objective To determine the protective effects of Chaihushugansan on endoplasmic reticulum(ER) in neuronal cells responding to the stress by using sero-pharmacological method.Methods The experiment used tunicamycin(TM) was induced in NG108-15 cells,the cells become ERS cell model,and Respectively with normal rabbit serum,Aricept rabbit serum containing high concentration,Chaihushugansan containing serum intervention.Reverse transcriptase PCR(RT-PCR) was utilized to detect the mRNA expressions of ubiquitin.Results The Chaihushugansan treated group at all teste d dosages showed significantly increase pxpressions with Tm treated control group(P〈0.05).Conclusions The Chaihushugansan has signifi cant protective effect for neuronal injury caused by endoplasmic reticulum stress,the mechanism may be associated with the ubiquitin-proteasome pathway.
