摘要
目的研究多壁碳纳米管(MWNTs)的光热效应及对人类肝癌HepG2细胞的杀灭作用。方法将多壁碳纳米管纯化、剪裁并用聚乙二醇(PEG)包覆,热电耦仪测定其光热效应。HepG2细胞分为对照组和3个剂量的MWNTs组,分别加入PBS和3个剂量(10,20,50μg·mL^(-1))MWNTs,与HepG2肝癌细胞共同培养24 h,在波长808 nm下,用红外光以强度2 W·cm^(-2)照射细胞5 min,用噻唑蓝比色法测定MWNTs对HepG2肝癌细胞的抑制作用;用AO-EB荧光染色测定MWNTs对HepG2肝癌细胞坏死的影响。结果在激光照射下,MWNTs迅速升温,温度可达56℃,与对照组相比,10μg·mL^(-1)MWNTs可明显抑制HepG2增殖(P<0.01),20μg·mL^(-1)MWNTs可杀灭HepG2细胞达80%以上,50μg·mL^(-1)MWNTs可杀灭HepG2细胞90%以上。结论多壁碳纳米管具有很强的光热转化效率,对HepG2肝癌细胞有强大的杀灭作用。
Objective To investigate the cytotoxic effect of multi-walled carbon nanotubes( MWNTs) on human liver HepG2 cells.Methods MWNTs were purified and coated with polyethylene glycol( PEG). Photothermal conversion efficiency of MWNTs was measured by thermocouple. The MWNTs at concentrations of 10,20 and 50 μg·mL^-1 were incubated with human liver HepG2 cells for 24 hours respectively and irradiated for 5 min at 880 nm at a laser power density of 2 W·cm^-2. The cell viability was evaluated by MTT assay and AO- EB fluorescent staining. Results MWNTs were coated evenly by PEG. The temperature of MWNTs was increased to 56 ℃ at 2 W · cm^-2laser power.HepG2 cells were killed nearly 80% with 20 μg · mL^-1 MWNTs and90% with 50 μg·mL^-1 MWNTs. Conclusion MWNTs had strong photothermal conversion efficiency and a strong inhibition to HepG2 cells.
