摘要
目的探索连翘苷对脂多糖(LPS)刺激的小胶质细胞BV-2的细胞炎症因子释放的抑制作用和机制。方法采用LPS(0.1 g/ml)刺激小胶质细胞建立神经退行性疾病(帕金森病)炎症模型,MTT法检测连翘苷对BV2的细胞毒性;硝酸还原酶法检测连翘苷对LPS刺激BV2细胞一氧化氮(NO)表达量的影响;一氧化氮合酶分型法检测连翘苷对LPS刺激BV2细胞一氧化氮合酶(i NOS)活性的影响;Western-blot法检测连翘苷对LPS刺激BV-2细胞TLR4蛋白表达影响。结果 50~200μg/ml连翘苷能抑制LPS诱导BV2细胞的炎症反应。结论连翘苷能够抑制小胶质细胞活化产生的炎症反应,其机制可能是通过抑制TLR4信号通路而发挥效用。
Objective To study the inhibitory effect of Forsythin on LPS-induced BV-2 cell and explore the underlying mechanism of this protective effect. Methods BV-2 cells were treated with LPS( 0. 1 μg / ml) for disease( Parkinson's disease,PD) inflammation model. MTT assay was used to detect the viability of BV-2 cells. NO was detected by the method of nitric acid reductase assay. i NOS enzyme activity was determined by type of nitric oxidesynthase assay. TLR4 protein expression was examined by the Western blot analysis. Results Forsythin did not exhibit cytotoxicity of BV2 cells.( 50 ~200 μg / ml) Forsythin significantly suppressed i NOS activity,the production of NO,i NOS and TRL4 protein expression in LPS-stimulated BV2 microglia cells in a dose dependent manner. Conclusion LPS induced activation of microglia lead to inflammatory response and its mechanism may be related to inhibiting TLR4 signaling pathway.
