摘要
为了探讨脂肪酸转运蛋白1(Fatty acid transport protein 1,FATP1)作为鸭屠体性状候选基因的可能性,试验以三穗鸭为材料,采用PCR-SSCP的方法进行单核苷酸多态性(SNPs)检测和基因型的分析。结果表明:在FATP1基因中检测到1个碱基变异,位于第5内含子65 bp处,为G→A突变,发现2种基因型(GA基因型、GG基因型),且在此位点的优势等位基因型为GA基因型,G为优势等位基因。在第1外显子、第2外显子、第4外显子、第5外显子和第8外显子上没有发现多态位点。关联分析结果表明,FATP1基因第5内含子处多态性对三穗鸭的全净膛、胸肌重、屠体重、腿肌重等性状存在显著影响(P<0.05)。
To investigate the possibility of fatty acid transport protein 1 ( FATP1 ) as a candidate gene of the slaughter traits in Sansui ducks, a polymerase chain reaction -single strand conformation polymorphism (PCR -SSCP) method was used for the detection of single nucleotide polymorphisms (SNPs) and analysis of genotypes, taking Sansui ducks as materials. The results showed that an G→A base mutation was detected in 65 bp of the intren 5 of FATP 1 gene, and two kinds of genotypes were found, including GA and GG; GA was a dominant allelic genotype at the site, and G was a dominant allele. No polymorphic sites were found in the exon 1, exon 2, exon 4, exon 5 and exon 8. The results of correlation analysis indicate that polymorphism in the intron 5 of FATP1 gene has a significant effect ( P 〈 0.05 ) on the existing traits of Sansaiducks, including eviscerated weight, breast muscle weight, leg muscle weight, carcass weight, and so on
