结直肠癌组织中Id-1、MMP-9的表达变化及其意义 被引量:2

Expression changes of Id-1 and MMP-9 in colorectal carcinoma tissues and its clinical significance

摘要 目的观察结直肠癌组织中癌基因DNA结合分化抑制蛋白1(Id-1)、基质金属蛋白酶9(MMP-9)的表达变化,并探讨其意义。方法结直肠癌组织50例份(观察组),正常结直肠组织50例份(对照组),分别采用免疫组化法、RT-PCR法、Western blotting法检测两组Id-1、MMP-9,分析Id-1、MMP-9阳性表达与结直肠癌临床病理参数的关系。结果观察组及对照组Id-1阳性表达率分别为72%、24%,MMP-9阳性表达率分别为78%、22%,两组比较,P均<0.05。Id-1、MMP-9阳性表达均与结直肠癌浆膜浸润、TNM分期、淋巴结转移、肝转移及脉管浸润相关(P均<0.05)。观察组及对照组Id-1 mRNA的相对表达量分别为0.96±0.03、0.20±0.04,MMP-9 mRNA的相对表达量分别为0.88±0.04、0.21±0.03,两组比较,P均<0.05。观察组及对照组Id-1蛋白的相对表达量分别为0.82±0.04、0.31±0.02,MMP-9蛋白的相对表达量分别为0.86±0.05、0.29±0.03,两组比较,P均<0.05。结论结直肠癌组织中Id-1、MMP-9表达升高,二者表达与结直肠癌浆膜浸润、TNM分期、淋巴结转移、肝转移及脉管浸润相关。 Objective To observe the expression changes of inhibitor of DNA binding-1 ( Id-1 ) and matrix metallo-proteinase-9 (MMP-9) in the human colorectal carcinoma tissues and to discuss the clinical significance. Methods The expression of Id-1 and MMP-9 in 50 specimens of colorectal carcinoma tissues ( observation group) and 50 specimens of normal colorectal tissues ( control group) was detected by immunohistochemistry, RT-PCR and Western blotting. The cor-relations between their positive expression and the clinicopathological features were analyzed. Results The positive rates of Id-1 in the observation group and the control group were 72% and 24%, the positive rates of MMP-9 were 78% and 22%, respectively, and there were significant differences between these two groups (all P〈0. 05). The positive expression of Id-1 and MMP-9 was related with the depth of tumor invasion, TNM stage, lymph node metastasis, vessel invasion and liver metastasis (all P〈0. 05). The relative expression of Id-1 mRNA in the observation group and the control group was 0. 96 ± 0. 03 and 0. 20 ± 0. 04, the relative expression of MMP-9 mRNA was 0. 88 ± 0. 04 and 0. 21 ± 0. 03, respectively, and there were significant differences (all P〈0. 05). The relative expression of Id-1 protein was 0. 82 ± 0. 04 and 0. 31 ± 0. 02, the relative expression of MMP-9 in the observation group and the control group was 0. 86 ± 0. 05 and 0. 29 ± 0. 03, respectively, and there were significant differences between these two groups (all P〈0. 05). Conclusion The expression of Id-1 and MMP-9 was increased in the human colorectal carcinoma tissues, and the expression was related with tumor in-vasion, TNM stage, lymph node metastasis, liver metastasis and vessel invasion.
