丁苯酞腹腔注射对大鼠局灶性脑缺血再灌注损伤的预防作用及其机制探讨 被引量:5

摘要 目的观察丁苯酞腹腔注射对大鼠局灶性脑缺血再灌注损伤的预防作用,并探讨其可能机制。方法60只成年健康SD大鼠随机分为A、B、C组各20只,A组大鼠腹腔注射400 mg/(kg·d)丁苯酞,B、C组予相同体积生理盐水,共1周。A、B组均制备局灶性脑缺血再灌注损伤模型,C组仅暴露颈总动脉及分叉处。比较各组大鼠神经行为学评分、脑梗死体积及脑组织凋亡细胞数、核转录因子κB(NF-κB)蛋白、NF-κB mRNA。结果与C组比较,A组和B组神经行为评分、脑梗死体积、脑组织凋亡细胞数及NF-κB蛋白、mRNA阳性细胞数增加;与B组比较,A组神经行为评分、脑梗死体积、脑组织凋亡细胞数及NF-κB蛋白、mRNA阳性细胞数减少;P均<0.01。结论丁苯酞对大鼠脑缺血再灌注损伤有预防作用,其机制可能与抑制NF-κB表达、减少脑组织凋亡细胞有关。
