HPLC同时测定布渣叶总黄酮提取物中6种黄酮类成分 被引量:8

Simultaneous Determination of Six Flavonoids in Extract of Microctis Folium by HPLC

摘要 目的:建立布渣叶总黄酮提取物中表儿茶素、牡荆苷、异牡荆苷、山柰酚-3-O-芸香糖苷、异鼠李素-3-O-葡萄糖苷和水仙苷等6种黄酮类成的HPLC同时测定方法。方法:采用HPLC法,Agilent HC-C_(18)色谱柱(4.6 mm×250 mm,5μm),甲醇-0.05%磷酸水溶液为流动相进行梯度洗脱,检测波长288 nm,柱温35℃,流速0.8 m L·min^(-1)。结果:表儿茶素、牡荆苷、异牡荆苷、山柰酚-3-O-芸香糖苷、异鼠李素-3-O-葡萄糖苷和水仙苷分别在131.200~1 312.000,149.800~1 348.200,131.400~1 051.200,99.175~595.050,109.955~659.730,267.000~1 348.200 ng呈线性关系,平均回收率分别为98.13%,97.41%,96.93%,98.22%,97.37%,97.82%。结论:该方法准确、可靠,重复性好,可用于布渣叶总黄酮提取物的质量评价与控制。 Objective: To establish a HPLC method for simultaneous determinationof the content of epicatechin, vitexin, isovitexin, kaemferol-3-O-rutinoside, isorhamnetin-3-O-β-D-glucoside, and narcissi in extract of Microctis Folium. Method: The HPLC analysis was carried out on Agilent HC-C18column( 4. 6 mm ×250 mm,5 μm) with amethanol-0. 05% phosphoric acid solution as the mobile phase for gradient elution at a flow rate of 0. 8 m L·min- 1. The detection wavelength was 288 nm,and the column temperature was 35 ℃. Result:Good linear relationship were found in the the range of 131. 200-1 312. 000 ng for epicatechin, 149. 800-1 348. 200 ng for vitexin, 131. 400-1 051. 200 ng for isovitexin, 99. 175-595. 050 ng for kaemferol-3-Orutinoside,109. 955-659. 730 ng for isorhamnetin-3-O-β-D-glucoside, and 267. 000-1 348. 200 ng for narcissi.Their average recoveries were 98. 13%, 97. 41%, 96. 93%, 98. 22%, 97. 37%, 97. 82%, respectively.Conclusion: The method is accurate,reliable,and can be used for the quality control and evaluationof total flavonoid extracts of Microctis Folium.
