蒙古族药冬青叶-7胶囊对平阳霉素致大鼠肺纤维化的治疗作用及机制分析 被引量:2

Efficacy and Mechanism of Mongolian Dongqingye-7 Capsule for Pulmonary Fibrosis Induced by Pingyangmycin

摘要 目的:观察蒙古族药冬青叶-7胶囊对平阳霉素致大鼠肺纤维化的治疗作用及初步分析作用机制。方法:90只健康SD大鼠,适应性饲养1周后随机分成假手术组和模型组,采用气管软骨环间隙注入平阳霉素方法建立大鼠肺纤维化模型。术后第7天分组,分别给冬青叶-7胶囊(120,240,480 mg·kg^(-1)),地塞米松组(0.3 mg·kg^(-1)),假手术组和模型组给予等量生理盐水。给药后第7,14,28天分别取血检测总抗氧化能力(T-AOC),取右肺检测转化生长因子-β1(TGF-β1)和羟脯氨酸(Hyp)含量,左肺固定。结果:第7,14,28天各观察点模型组与假手术组比较TGF-β1蛋白表达均明显上升(P<0.05,P<0.01);肺系数明显升高(P<0.01);肺组织之中的Hyp含量明显升高(P<0.05,P<0.01);血清总抗氧化能力明显降低(P<0.05,P<0.01);各剂量组与模型组比较TGF-β1蛋白表达:蒙族药各剂量组明显减弱(P<0.05,P<0.01),激素组14,28 d有明显减弱(P<0.05);肺系数:冬青叶-7胶囊各剂量组和激素组均明显降低(P<0.05,P<0.01);肺组织之中的Hyp含量:冬青叶-7胶囊高剂量组和激素组7,14,28 d均有明显降低(P<0.05,P<0.01),中、低剂量组14,28 d明显降低(P<0.05,P<0.01);血清总抗氧化能力中、冬青叶-7胶囊高剂量组和激素组14,28 d明显增高(P<0.05,P<0.01),低剂量组28 d明显增高(P<0.01)。结论:蒙古族药冬青叶-7胶囊对平阳霉素致大鼠肺纤维化有一定的治疗作用,其机制初步分析可能与TGF-β1的表达有关。 Objective: To observe the therapeutic effect and mechanism of Mongolian Dongqingye-7capsule for the pulmonary fibrosis induced by pingyangmycin in rats. Method: Totally 90 healthy SD rats were randomly divided into sham-operation group and model group after 1 week of adaptive breeding. Gap injection of pingyangmycin was done by using tracheal cartilage ring to establish the pulmonary fibrosis models in rats.Postoperative day 7,the rats were divided into Mongolian Dongqingye-7 capsule groups( 120,240,480 mg·kg-1),dexamethasone group( 0. 3 mg·kg-1),sham-operation group and model group with same amount of normal saline. 7,14,28 days after the treatment,blood was taken to detect total antioxidant capacity( T-AOC); right lung was taken to detect transforming growth factor-β1( TGF-β1) and hydoroxyproline( Hyp) content,while the left lung was fixed. Result: On day 7,14,28,as compared with the sham-operation group,the TGF-β1protein expressions were significantly increased( P〈0. 05,P〈0. 01); lung coefficient was increased significantly( P〈0. 01); Hyp content in lung tissues was increased significantly( P〈0. 05, P〈0. 01); and total antioxidant capacity in serum was decreased significantly( P〈0. 05,P〈0. 01) in the model group at each observation point.As compared with the model group, TGF-β1protein expression was decreased significantly in Mongolian drug groups( P〈0. 05,P〈0. 01),and significantly decreased on day 14 and day 28 in hormone group( P〈0. 05);lung coefficient was significantly decreased in both Mongolian drug groups and hormone group( P〈0. 05,P〈0. 01); Hyp content in liver tissues was significantly decreased on days 7,14,28 in Mongolian drug high dose group and the hormone group( P〈0. 05,P〈0. 01),and significantly decreased on days 14 and 28 days in Mongolian drug low dose group and mid dose group( P〈0. 05,P〈0. 01); total antioxidant capacity in serum was significantly increased on day 14 and day 28 in Mongolian drug mid dose group,high dose group and hormone group( P〈0. 05,P〈0. 01),and significantly increased on day 28 in Mongolian drug low dose group( P〈0. 01). Conclusion: Mongolian Dongqingye-7 capsule has certain therapeutic effects for the pulmonary fibrosis induced by pingyangmycin in rats,and its preliminary analysis shows that the mechanism may be related to the expression of TGF-β1.
