摘要
研究了猴头菇多糖协同ConA对小鼠体外脾淋巴细胞分泌Th1、Th2细胞因子的量及mRNA表达量的影响,旨在探讨其协同免疫调节机制。用ELISA法检测HEP协同ConA刺激脾淋巴细胞细胞上清液中Th1、Th2细胞因子的量;RT-PCR法检测Th1、Th2细胞因子和转录因子mRNA的表达。结果显示,HEP在25-400mg/L质量浓度范围内能显著协同ConA刺激T细胞亚群Th1细胞因子(IL-2、IFN-γ、TNF-α)和Th2细胞因子(IL-4、IL-6)的分泌及mRNA的表达(P〈0.05),并显著促进转录因子(T-bet、Gata-3)mRNA的表达(P〈0.05)。结果表明,HEP能协同ConA促进小鼠脾淋巴细胞Th1、Th2细胞因子的分泌及基因的表达,通过调节Th1/Th2平衡,发挥双重免疫调节作用。
To investigate the regulating effects of polysaccharides from Hericium erinaceum on Th1,Th2 cytokine secretion and gene expression in murine splenic lymphocyte induced by ConA.The contents of Th1,Th2 type cytokines of murine splenic lymphocyte treated with ConA and different concentration of HEP were determined by ELISA.The gene expression of Th1,Th2 type cytokines and transcription factors were determined by RT-PCR.HEP at a concentration ranging from 25-400 mg/L could elevate the levels and gene expression of Th1cytokines(IL-2,IFN-γ,TNF-α)and Th2cytokines(IL-4,IL-6)after sensitization with ConA.HEP also could elevate the mRNA expression of transcription factors(T-bet,Gata-3).HEP could promote the secretion and mRNA expression of Th1,Th2 type cytokines in murine splenic and elicit a balance Th1/Th2 response increase both cellular and humoral immune responses
