摘要
目的:应用银芍散干预高脂饮食诱导的单纯性脂肪肝模型大鼠,明确银芍散对非酒精性单纯性脂肪肝的防治作用。方法:采用高脂饲料加10%果糖水喂养18周,诱导SD大鼠形成单纯性脂肪肝,造模同时灌胃给药,记录体重,观察一般行为学。末次给药后取下腔静脉血和肝脏,检测血清血脂四项(TC、TG、HDL-c、LDLc)及肝功能指标(ALT、AST)。计算肝指数,HE染色观察肝脏组织病理的变化,并检测肝组织中总脂酶活性和FFA水平。结果:与正常组相比,模型组大鼠体重增长较为明显,血清TC、TG、LDL-c、ALT、AST、肝指数及肝中FFA水平明显升高,血清HDL-C水平及肝总脂酶活性明显降低(P<0.01,P<0.05)。病理学显示模型组大鼠出现了严重脂肪变和不同程度炎症细胞浸润、汇管区渗出、肝细胞坏死。应用银芍散干预后,大鼠血清TC、TG、LDL-c、ALT、AST、肝指数及肝中FFA水平明显降低,血清HDL-C及肝总脂酶活性明显升高(P<0.01,P<0.05)。同时可明显减轻大鼠肝肿大,改善肝细胞的脂肪变性和炎症程度。结论:银芍散可通过纠正血脂紊乱,改善肝功能和肝细胞脂肪变性防治单纯性脂肪肝,其作用机制可能与提高肝总脂酶活性,减少肝中FFA,进而增加TG分解,降低TG合成有关。
Objective: Yinshao San( YSS) was applied in intervention of the non- alcoholic simple fatty liver( SFL)model rats induced by high- fat diet in order to define the effect on the prevention and treatment of non- alcoholic simple fatty liver by YSS. Method: High fat feeding with drinking 10% fructose was used to set up SFL model in SD rats. And meanwhile give drugs by intragastric administration,record the weight and observe behavior in general in 18 consecutive weeks. Blood in inferior vena cava and liver from SD rats was taken out after the last administration. Serum lipid parameters( TC,TG,HDL- C,LDL- C) and liver function indexes( ALT,AST) were tested at last week. Observe the liver tissue pathological changes by HE staining and test the HL,LPL,FFA level in liver tissue in SFL rats. Result: Compared with the normal control group,weight gain in model group was more obvious. The content of TC,TG,LDL- C,ALT and AST in serum and the FFA level in liver tissue in model control group has been significantly increased. The content of HDL- C in serum and the HL and LPL activity in liver tissue in model control group were significantly decreased( P〈0. 01,P〈0. 05). In addition,the histopathology showed increased fatty dropletswithin hepatocytes and liver inflammation. After applying YSS intervention,the contents of TC,TG,LDL- C,ALT and AST in serum and the FFA level in liver tissue in model control group were significantly decreased. The contents of HDL- C in serum and the HL and LPL activity in liver tissue in model control group were significantly increased( P〈0. 01,P〈0. 05). At the same time,it can significantly reduce rats hepatomegaly,improve steatosis and inflammation of liver cells. Conclusion: YSS can prevent simple fatty liver,primarily by correcting dyslipidemia and improving liver function and hepatic steatosis. The mechanism may in-crease hepatic lipase activity and reduce liver FFA,and thus increasing TG decomposition and reducing TG synthesis.
