复方钩藤降压片对SHR腹主动脉壁白细胞黏附浸润及E-选择素表达的影响

Compound Gouteng Jiangya Tablet on Leukocyte Adhesion,Infiltration and E-Selectin Expression in SHR Abdominal Aortic Wall

目的:试从干预高血压病血管壁炎症细胞浸润角度,进一步完善复方钩藤降压片降压及防治靶器官损害的机制。方法:21只7周龄雄性自发性高血压大鼠(SHR)随机分为3组:模型组(SHR-S组)、复方钩藤降压片组(SHR-G组)、缬沙坦组(SHR-D组),另7只WKY大鼠为空白对照组(WKY组)。连续干预6周,HE染色观察腹主动脉白细胞黏附跨壁浸润情况、免疫组化法测定管壁E-选择素(Es)的表达水平。结果:(1)对腹主动脉壁白细胞黏附浸润的影响:SHR-S组管壁皆有不同程度白细胞黏附浸润,以黏附为主,内皮连续性较差,弹性纤维排列紊乱,平滑肌细胞显影模糊,部分大鼠平滑肌细胞有空泡样变性,SHR-D组管壁均有不同程度的白细胞黏附浸润,以中膜浸润为主,未见其他明显异常改变,WKY组及SHR-G组动脉管壁无上述异常病变;(2)对腹主动脉Es表达的影响:Es的表达水平SHR-G组低于SHR-S组(P<0.05),与WKY组与SHR-D组无显著差异(P>0.05),SHR-D组表达水平亦低于SHR-S组(P<0.05)。结论:SHR腹主动脉壁存在白细胞黏附浸润,且E-选择素表达水平升高,复方钩藤降压片能有效减少管壁白细胞黏附浸润及降低E-选择素的表达水平。

Objective： From the intervention of inflammatory cell infiltration in hypertensive vascular wall,further to improve the mechanism of Compound Gouteng Jiangya Tablet in lowering blood pressure and preventing target organ damage. Methods： A total of 21 SHR rats（ 7 weeks old,male） were randomly divided into 3 groups： Spontaneously hypertensive rats model group（ Group SHR- S）,Fufang Gouteng Tablets group（ Group SHR- G）,Valsartan group（ Group SHR- D）. Another 7 WKY rats were the normal control group（ Group WKY）. After 6 weeks treatment,HE staining was used to observe the abdominal aortic wall leukocytes infiltration and adhesion and immunohistochemical detection for Es expression in abdominal aorta. Results：（ 1） Effect on leukocyte adhesion and infiltration： SHR- S group had different degrees of leukocyte infiltration and adhesion. The endothelialcontinuity was poor and the arrangement of elastic fiber disordered and smooth muscle cells vague. Parts of smooth muscle cells had free bubble- like degeneration. SHR- D group＇s white blood cells adhesion wall had different degrees of infiltration,mainly infiltration in media. And others had no abnormal changes. WKY group and SHR- G group had no abnormal lesions in the arterial wall.（ 2） Effect on the expression of Es： The expression level of SHR- G was lower than that of the group SHR- S（ P〈0. 05） and group WKY had no significant difference compared with the SHR- D group（ P〉0. 05）. The expression level of SHR- D group was lower than that of group SHR- S（ P〈0. 05）. Conclusion： SHR abdominal aortic wall has leukocyte adhesion and infiltration and the expression of Es is high. Compound Gouteng Jiangya Tablet can effectively reduce it.
