摘要
目的:通过醋酸致大鼠溃疡性模型试验,探讨久泻宁颗粒治疗急性溃疡性结肠炎的机制。方法:除空白组外,其他各组以2,4-二硝基氯苯和醋酸联合应用建立大鼠溃疡性结肠炎模型。造模成功后,将70只SD大鼠分层随机分空白组、UC模型对照组、补脾益肠丸阳性对照组、柳氮磺胺吡啶片阳性对照组、久泻宁颗粒高剂量组、中剂量组、低剂量组,每组10只(雌雄各半)。空白组与UC模型对照组给饮水,其他分别给相应的药物,连续14天后,腹主动脉采血测定:血清IL-2、TNF-α、免疫球蛋白Ig G、Ig M以及SOD、MDA,同时取结肠溃疡明显处作病理分析。结果:UC模型对照组与正常组比较,UC模型对照组明显出现免疫异常(P〈0.01),血清TNF-α、Ig G、Ig M水平明显升高(P〈0.01),IL-2水平明显降低(P〈0.01)。与模型对照组比较,久泻宁颗粒组TNF-α、Ig G、Ig M、MDA水平降低(P〈0.05~P〈0.01),IL-2、SOD水平升高(P〈0.05~P〈0.01)。结论:久泻宁颗粒能明显调节UC大鼠机体免疫功能异常,对模型大鼠结肠溃疡面有较好的修复和恢复作用。
Objective: According to acetic acid- induced ulcer model of rats,to investigate Jiuxiening Particle treatment mechanism of acute ulcerative colitis. Method: Expert the control group,2,4- dichlorobenzene and acetic acid nitro were used to establish the rat model of ulcerative colitis. After the modeling,the hierarchical 70 SD rats were randomly divided into blank,UC model group,Bupi Yichang Pill positive control group,Sulfasalazine Tablets positive control group,Jiuxiening Particles high dose group,middle dose group,low- dose group( 10 in each,half male and half male). The blank control group and UC model group were given water and the others given appropriate medication for 14 consecutive days. Abdominal aortic blood assay: serum IL- 2,TNF- α,immunoglobulins Ig G,Ig M and SOD,MDA,while taking colon ulcer obvious place for histological analysis. Results: UC model group obviously appeared abnormal immune and serum TNF- α,Ig G and Ig M levels were significantly increased( P〈0. 01) and IL- 2 levels were significantly lower( P〈0. 01),combined with rats bowel condition and daily symptoms and pathological analysis of colon tissue,indicating the successful model. Compared with the model group,Bupi Yichang Pill group and sulfasalazine group after oral administration,Jiuxiening Particle group can significantly reduce TNF- α,Ig G,Ig M and MDA levels( P〈0. 05,P〈0. 01) and elevate IL- 2 and SOD levels( P〈0. 05,P〈0. 01). Conclusion: 1. Jiuxiening Particle can regulate UC immune dysfunction in rats and colon ulcers in rats can be better repaired and the mechanism may be regulating the immune function to weaken their immune response.
