摘要
目的探讨Polo样蛋白激酶4(PLK4)表达与子宫内膜异位症(EMS)的相关性。方法 2013年5月至2014年1月在浙江大学医学院附属妇产科医院,采用免疫组织化学、实时荧光定量PCR技术(Real-time q PCR)以及蛋白免疫印迹的方法对37例异位子宫内膜(异位组)标本及32例正常子宫内膜(正常组)标本中PLK4的表达情况进行检测。结果免疫组织化学提示PLK4在异位组中的表达明显高于正常组,且差异有统计学意义(P<0.05)。PLK4 m RNA在异位组及正常组中均有表达,但异位组中的相对表达量(0.01190±0.00574)明显高于正常组(0.00227±0.00120),差异有统计学意义(P<0.01)。蛋白免疫印迹提示,PLK4蛋白在正常组及异位组内膜组织中均有表达,但异位组PLK4蛋白的表达显著高于正常组(P<0.01)。结论 PLK4在异位内膜组织中的高表达可能导致异位内膜加速生长,可能与EMS在临床上所表现出的恶性生物学行为有一定的关联。
Objective To explore the expression of PLK4 in the endometriosis (EMS), and analyze the correlation of PLK4 with endometriosis (EMS). Methods The ectopic endometria samples were collected including 37 EMS patients, and 32 normal endometria samples without endometriosis from May. 2013 to Jan. 2014 in Women's Hospital, School of Medicine, Zhejiang University.The expressions of PLK4 were examined by immunohistochemistry, real-time qPCR and Western Blot respectively, then analyzed statistically.Results According to immunohistochemistry, the expression of PLK4 in ectopic endometrium tissues was significantly higher than that in normal endometrium tissues (P 〈 0.05).Real- time qPCR showed that the expression level of PLK4 mRNA in ectopic endometrium tissues was significantly higher than that in normal endometrium tissues (0.01190 ±0.00574 vs. 0.00227 ± 0.00120) , P 〈 0.01 ).Furthermore, western Blot showed that the expression of PLK4 protein could be detected both in ectopic and normal endometrium tissues, but the expression in ectopic endometrium tissues was significantly higher than that in normal endometrium tissues (P 〈 0.01 ). Conclusion The high expression of PLK4 may promote the growth of the ectopic endometria of endometriosis, and may correlate with the malignant biological behaviors of EMS.
