摘要
目的观察天麻素对脑缺血再灌注大鼠脑组织中缺氧诱导因子-1α(HIF-1α)、血管内皮生长因子(VEGF)等基因mRNA表达的影响。方法灌胃不同剂量天麻素7 d,采用线栓法制备大鼠大脑中动脉局灶性脑缺血(2 h)再灌注损伤模型,造模成功后继续给药7 d。采用实时荧光定量PCR(Real Time RT-PCR)法检测缺血半暗带脑组织中HIF-1α、VEGF、BDNF、MMP2、e NOS、PPARα及SOD1 mRNA的表达。结果天麻素可上调HIF-1α、VEGF、BDNF、MMP2、e NOS、PPARα、SOD1 mRNA的表达(P<0.05),下调MMP2 mRNA的表达(P<0.05)。结论天麻素可通过上调大鼠脑缺血半暗带HIF-1α、VEGF、BDNF、MMP2、e NOS、PPARα、SOD1 mRNA的表达,下调MMP2 mRNA的表达,产生抗局灶性脑缺血再灌注损伤的作用。
Objective To explore the effects of Gastrodin( Gas) on the mRNA expressions of hypoxia inducible factor- 1α( HIF- 1α),vascular endothelial growth factor( VEGF) and other genes in the ischemic penumbra of rats injured by ischemia( 2 h)- reperfusion( I / R). Methods Rats were pretreated with Gas for 7 days followed by the focal cerebral I / R and then Gas was withdrawn 7 days after the I / R surgery. The mRNA expressions of HIF- 1α,VEGF,brain derived neurotrophic factor( BDNF),matrix metalloproteinase- 2( MMP2),endothelial nitric oxide synthase( e NOS),peroxisome proliferators activated receptors( PPARα) and superoxide dismutase- 1( SOD1) in the ischemic penumbra of rats were detected by Real Time RT- PCR. Results The expressions of HIF- 1α,VEGF,BDNF,MMP2,e NOS,PPARα and SOD1 mRNA in Gas group increased obviously( P 〈 0. 05),while the expression of MMP2 mRNA in Gas group decreased obviously( P 〈 0. 05).Conclusion Gas has protective effects on I / R- injured rat brain by up- regulating the expressions of HIF- 1α,VEGF,BDNF,e NOS,PPARα and SOD1 and down- regulating the expression of MMP2.
