穿心莲内酯对心肌肥厚大鼠心肌胶原及心肌酶谱的影响

Anti- cardiac hypertrophy effect of andrographolide on myocardial collagen and myocardial enzymes

目的研究穿心莲内酯对心肌肥厚大鼠心肌胶原及心肌酶谱的影响。方法健康雄性SD大鼠随机分为对照组,心肌肥厚模型组,溶剂对照组,低剂量,高剂量穿心莲内酯组。采用连续10天背部皮下注射异丙肾上腺素建立心肌肥厚模型。连续给药治疗14 d后,心脏取血,分离血清,测定心肌酶谱(LDH、CK、SOD、MDA)和心肌胶原Collage-Ⅰ的变化。结果穿心莲内酯能显著降低心肌胶原Collage-Ⅰ的含量;能显著降低血清LDH、CK活性及MDA含量;能显著升高SOD活性。结论穿心莲内酯能通过降低心肌胶原Collage-Ⅰ的含量及改变心肌酶谱,从而达到抑制心肌细胞肥厚作用。

Objective To study the anti- cardiac hypertrophy effect of andrographolide on myocardial collagen and myocardial enzymes. Methods Healthy male SD rats are randomly divided into five groups including control group,cardiac hypertrophy model group,solvent control group,low- dose andrographolide group,and high- dose andrographolide group. The cardiac hypertrophy model was set up by subcutaneous injection of isoproterenol for 10 consecutive days. After treated for another 14 days,the rats in treating groups,as well as in other groups,were sacrificed and cardiac punctured,serum was separated and determined for the variation of LDH,CK,SOD,MDA and myocardial collagen Collage- I. Results Andrographolide significantly decreased the content of myocardial collagen Collage- I,together with the activities of LDH and CK,the content of MDA in serum. The SOD activity was significantly increased as well. Conclusion Andrographolide inhibits cardiac hypertrophy by decreasing the myocardial collagen Collage- I content and regulating myocardial enzymes.
