针灸对骨髓抑制小鼠DNA修复基因XPD蛋白表达的影响 被引量:16

摘要 目的通过测定针刺和艾灸前后,骨髓抑制小鼠DNA修复基因XPD蛋白表达量的改变,探讨针灸减轻骨髓抑制、增加外周血白细胞的分子生物学机制。方法取清洁级、雄性昆明种小鼠224只,分成正常组、模型组、针刺组、艾灸组。模型组、针刺组、艾灸组的小鼠腹腔注射环磷酰胺(CTX),造成CTX骨髓抑制小鼠模型,正常组小鼠腹腔注入等量的0.9%氯化钠溶液。针刺组、艾灸组分别进行针刺、艾灸,正常组、模型组陪同固定,不治疗。各组均在2~6d用免疫组化染色法(Elivison二步法)、Western Blot法检测和分析骨髓细胞XPD蛋白表达量的变化。结果针灸可以通过上调CTX模型小鼠骨髓细胞中XPD的蛋白表达,促进骨髓细胞受损DNA的核苷酸切除修复,减轻因CTX化疗引起的骨髓抑制。结论上调受损骨髓细胞DNA修复基因XPD的表达,促进细胞受损DNA核苷酸切除修复,是针灸减轻骨髓抑制、保护造血功能、增加外周血白细胞的重要机制之一。 Objective By observing the effects of acupuncture and moxibustion on expression of protein of DNA repair Gene XPD in the bone marrow inhibition's mice,to explore the molecular biological mechanism that acupuncture and moxibustion can relieve myelosuppression and increase peripheral blood leukocytes. Methods Dividing the 224 clean male Kun- ming mice into control,model,acupuncture and moxibustion groups randomly. The later three groups were made the model with cyclophosphamide( CTX)intraperitoneal injection,while the control with equal injection of 0. 9 % of the saline solution. The acupuncture and moxibustion groups were respectively used for treatment with acupuncture and moxibustion,while the control and model group were caught and fixed without treatment. Between groups in 2nd to 6th day with the immunohistochemical staining method( Elivison on two steps),Western Blot method to detect and analyze the quantity of bone marrow cells XPD protein expression. Results Acupuncture and moxibustion can increase XPD protein expression in bone marrow cells in model of CTX mice,promote the nucleotide excision repair and antagonism alkylating agent toxicity caused by CTX. Conclusion Acupuncture and moxibustion can relieve bone marrow suppression,protect hematopoietic function,increase the one of the important mechanism of the peripheral blood leukocytes. One of its main mechanisms is increasing the expression of DNA repair gene XPD of bone marrow cells,promoting cells damaged DNA nucleotide excision repair.
