摘要
目的分析棕榈酸处理对小鼠神经母细胞瘤细胞(N1E-115)食欲相关肽神经肽Y(NPY)、刺鼠基因相关蛋白(AgRP)基因表达的影响,探讨脂肪酸转位酶(FAT/CD36)在此过程中的作用。方法取对数生长期的细胞用10μmol/L棕榈酸培养5、10、15、20min,牛血清白蛋白(BSA)为对照组,提取mRNA,检测细胞NPY和AgRP mRNA的表达。采用10μmol/L棕榈酸培养20 min,比较加或不加FAT/CD36抑制剂N-油酰基硫代琥珀酰亚胺(SSO)预处理对细胞NPY、AgRP、FAT/CD36mRNA表达的影响。结果 110μmol/L棕榈酸处理细胞,NPY、AgRP mRNA的表达随时间逐渐降低,20min降至最低;210μmol/L棕榈酸处理20min,加入SSO预处理30min组与未加入SSO组相比,NPY、AgRP mRNA表达降低的幅度明显减小。结论培养的N1E-115神经细胞中加入棕榈酸处理可抑制NPY、AgRP mRNA的表达,而这种抑制作用是通过FAT/CD36途径实现的。
Objective To explore the effect of palmitic acid on the gene expression of neusopeptide Y(NPY) and agouti gene-related protein(AgRP)in murine neuroblastoraa cell line N1E-115, and to investigate the role of fatty acid translocase (FAT/CD36)in the process. Methods N1E-115 cells at the logarithmic phase were treated with 10 μmol/L palraitic acid for 5, 10,15 and 20 min, with bovine serum albumin(BSA)serving as control. The expression of NPY and AgRP mRNA was detected in the cells. Additionally, the effect of sulfosucciniraidyl-oleate(SSO), an inhibitor of FAT/CD36, on the expression of NPY, AgRP and FAT/CD36 was examined in N1E-115 ceils treated with 10μmol/L palmitic acid for 20 min. Results OPalmitic acid (10 μmol/L)could significantly decrease the expression of NPY and AgRP mRNA in a time-dependent manner. ②The decrease in the expression of NPY and AgRP raRNA was significantly attenuated in cells treated with 10μmol/L palmitic acid for 20 min and SSO for 30 min. Conclusion Palraitic acid can suppress the expression of NPY and AgRP genes in NIE-115 ceils via the fatty acid transport protein FAT/CD36 pathway.
