补阳还五汤对脑出血大鼠脑组织海马区CXCR4-PI3K生物学自噬轴的作用 被引量:1

摘要 目的探讨补阳还五汤对脑出血大鼠CXCR4-PI3K自噬轴的影响及其机制。方法健康SD雄性大鼠72只按照Rosenberg法复制脑出血大鼠模型,随机分为假手术组、模型组、补阳还五汤组、银杏叶片组,每组各18只,补阳还五汤组、银杏叶片组分别采用补阳还五汤、银杏叶片灌胃,假手术组及模型组给予等剂量的生理盐水。采用免疫组化法(ELISA)检测PI3K、AKT、SDF-1α、CXCR4蛋白,TUNEL法检测细胞凋亡。结果与模型组比较,补阳还五汤组和银杏叶片组PI3K、AKT蛋白表达及SDF-1α、CXCR4阳性细胞数显著增高(P<0.05);模型组、补阳还五汤组、银杏叶片组大鼠TUNE阳性细胞较假手术组显著增加(P<0.05);补阳还五汤组和银杏叶片组TUNE阳性细胞较模型组显著减少(P<0.05)。结论补阳还五汤能够激活CXCR4-PI3K自噬轴,既能够激发细胞自噬,又能够调控细胞自噬状态,实现抑制细胞凋亡的脑保护作用。 Objective To explore the effect and mechanism of Buyanghuanwu Decoction on cxcr4 -PI3K autophagy pathway in cerebral hemorrhage rats.Methods According to Rosenberg method cerebral hemorrhage rat model was replicated,nd Buyanghuanwu Decoction gavage was administered only on rats in Group of Buyanghuanwu Decoction.lmmunohistochemistry method was used to detect the PI3K,Akt, SDFle, CXCR4 protein, cell apoptosis was detected by TUNEL method.Results Compared with model group, PI3K,and AKT protein expression and alpha SDF-1 and CXCR4 positive cells in Buyanghuanwu Decoction group and ginkgo leaf group were signifi- cantly higher (P〈0.05);rats TUNE positive cells in model group, Buyanghuanwu Decoction group and ginkgo leaf group increased significantly when compared with those in control group (P〈0.05) ;TUNE positive cells in Buyanghuanwu Decoction group and ginkgo leaf group de- creased significantly when compared with those in the model group (P〈0. 05).Conclusion Buyanghuanwu Decoction can activate the cxcr4-PI3K autophagy pathway to the effects that it not only activates autophagy, but also regulates autophagy state to inhibit apoptosis of cerebral pro tective effect.
