摘要
为了解犬流感病毒(CIV)在广州地区的流行和进化情况,本研究从广州市宠物医院采集了230份犬鼻咽拭子样品,接种10日龄SPF鸡胚,进行CIV的分离和鉴定,并对其全基因组序列进行分析。结果表明,从这些样品中仅分离鉴定出一株H3N2亚型CIV A/canine/Guangdong/01/2014(H3N2)。对该病毒株全基因组的遗传进化分析表明,分离株的PB2、PB1、PA、NP、HA、NA、M、NS 8个基因均为CIV,未发生重组;HA和NA基因与我国广东地区分离的H3N2亚型CIV属于同一个分支,氨基酸同源性分别达98.2%和97.0%。本研究结果为明确华南地区CIV的流行进化情况提供了基础数据。
To investigate the epidemic and variation of canine influenza virus (CIV) in Guangzhou area, a total of 230 nasal swabs from dogs were collected. A virus was isolated by inoculation in 10-day-old SPF embryonated eggs, and the virus was identified as H3N2 subtype by RT-PCR, named A/canine/Guangdong/01/2014 (H3N2). The complete genes were sequenced, phylogenetically analyzed and compared with the sequences of reference viruses. All of eight segments of the isolate were originated from H3N2 subtype CIV without recombination. In addition, the amino acid homology of HA and NA gene is up to 98.2% and 97.0%, respectively. This study provided valuable basic data for the epidemics and evolution of CIV in South China.
