摘要
基于硫酸卡那霉素与茚三酮作用形成的络合物在563nm处有特征吸收,以茚三酮为显色剂,采用可见分光光度法测定硫酸卡那霉素的含量。通过考察缓冲溶液的酸度及其用量、茚三酮溶液的用量、加热反应时间和加样顺序等条件得到最佳显色反应条件。硫酸卡那霉素的质量浓度在0.074~0.465g·L^(-1)范围内与吸光度呈线性关系,检出限(0.01/k)为0.026g·L^(-1)。加标回收率在95.5%~98.4%之间,测定值的相对标准偏差(n=12)小于3.0%。
Visible spectrophotometry was applied to the determination of kanamycin sulfate with ninhydrin as chromogenic agent,based on the chromogenic reaction of kanamycin sulfate with ninhydrin to form a complex with characterized adsorption at 563 nm.The optimum conditions for chromogenic reaction were defined by investigating the conditions including the acidity and dosage of buffer solution,the dosage of ninhydrin solution,reaction time through heating and sample order. A linear relationship was found between the absorbance and the mass concentration of kanamycin sulfate in the range of 0.074-0.465g·L-1 with detection limit(0.01/k)of 0.026g·L-1.Recovery rates obtained by standard addition method were in the range of 95.5%-98.4%,and RSDs(n=12)were less than 3.0%.
