基质金属蛋白酶-2,3在退变颈椎间盘髓核组织中的表达及意义 被引量:7

摘要 目的通过观察退变颈椎间盘髓核组织基质金属蛋白酶-2(MMP-2)和MMP-3表达的变化,探讨其在颈椎间盘退变过程中的作用及意义。方法 55例颈前路椎间盘摘除术的颈椎病患者髓核标本作为实验组,30例颈椎间盘切除术的颈椎外伤患者髓核标本作为对照组。将取出的标本固定后切片染色,采用免疫组化方法对标本内MMP-2和MMP-3染色阳性的细胞进行定性和半定量分析.观察其表达变化。结果实验组髓核细胞和软骨细胞的胞浆中MMP-2和MMP-3染色阳性率分别为78.18%和70.91%,明显高于对照组(P<0.05)。结论 MMP-2和MMP-3在退变颈椎间盘中高表达,二者可能在颈椎间盘退变巾发挥了一定作用。 Objective To study the expressions and significance of matrix metalloproteinase-2 (MMP-2) and matrix metalloprotcinase-3 (MMP-3) in nucleus pulposus of degenerative cervical intervertebral disc. Methods 55 patients with cervical spondylosis underwent anterior cervical discectomy and taken as experi- mental group. While 30 patients with cervical spine trauma received cervical discectomy as control group. Obtained specimens were fixed, sectioned and stained, the expression levels of the MMP-2 and MMP-3 were determined qualitatively and semiquantitatively by immunohistochemical methods. Results In the experimental group, the positive rates of MMP-2 and MMP-3 staining in the cytoplasm of nucleus pulposus cells and chon- drocytes were 78.18% and 70.91%, respectively, which were significantly higher than those in control group (P 〈 0.05). Conclusion It suggested that MMP-2 and MMP-3 play a role in the degeneration of cervical intervertebral disc.
