参芍口服液对2型糖尿病大鼠肝损害的干预作用 被引量:2

THE INTERVENTION OF SHENSHAO ORAL LIQUID ON TYPE 2 DIABETES-INDUCED LIVER INJURY

摘要 目的观察参芍口服液对2型糖尿病大鼠肝损害的干预作用。方法将30只大鼠随机分为对照组(C组)、模型组(M组)和治疗组(T组)。用链脲佐菌素制备2型糖尿病大鼠模型,12周后处死大鼠取血和肝组织。全自动生化法检测GLU、ALT、AST、TC、TG、LDL、CRP;免疫组化观察IL-6表达的变化。HE染色观察肝组织病理形态学改变;电镜观察各组大鼠肝脏的超微结构改变。结果与对照组比较,模型组GLU、ALT、AST、TC、TG、LDL、CRP显著增高(P<0.05);免疫组化显示:IL-6表达含量明显升高;HE显示:肝细胞弥漫性肿胀,水样变性,窦间隙狭窄;透射电镜可见肝细胞结构不清晰,胞质疏松,线粒体肿大,模糊,嵴断裂,粗面内质网破裂、排列紊乱,脂滴增多。经参芍口服液干预后,大鼠肝功能、血脂,肝组织形态学,肝脏超微结构均明显好转。结论参芍口服液对糖尿病肝损害大鼠有一定的保护作用,其机制可能是通过抑制炎症反应实现的。 Objective To explore the intervention of Shenshao oral liquid on type 2 diabetes- induced liver injury. Methods Thirty rats were randomly distributed into the control group(C), model group(M) and treatment group(T). A type 2 diabetic rat model was developed by administrating Streptozotocin. Twelve weeks later, the rats were sacrificed; blood samples and hepatic tissue were obtained. Full automatic bio- chemical analytic method was used to measure the level of GLU, ALT, AST, TC, TG, LDL, and CRP. Immun0histochemical staining was used to observe the expression of IL - 6 ; HE staining was applied to ob- serve morphology change on liver tissue. The ultrastructural changes of liver in rats were observed by elec- tron microscope. Results In group M, the level of GLU, ALT, AST, TC, TG, LDL, CRP were signifi- cantly increased ( P^0.05}. Immunohistochemical staining showed that the expression of IL - 6 increased significantly ; HE displayed that diffuse swelling of the liver cells, degeneration of water samples, and sinus stenosis. Electron microscopy showed that the structure of the liver cell was not clear, cytoplasm rarefac- tion and swelling of mitochondria, fuzzy, cristae fracture, rough endoplasmic reticulum rupture, arranged in disorder, lipid droplets was increased. After Shenshao oral liquid intervention, liver function, blood lip- id, liver morphology and ultrastructure of liver were significantly improved. Conclusion Shenshao oral liq- uid exhibits a protective effect against diabetes- related liver injury and may be achieved by inhibiting the inflammatory response.
