西藏尼玛县盐碱地嗜碱芽胞杆菌资源采集与鉴定 被引量:3

摘要 为调查西藏尼玛县盐碱地环境芽胞杆菌资源,采用纯培养法从西藏尼玛县采集的盐碱地样品中分离芽胞杆菌,利用16SrRNA基因进行系统发育进化分析。同时,选取代表性菌株进行生理生化特性分析。结果筛选到了14株芽胞杆菌,经16SrRNA基因鉴定均属于嗜碱芽胞杆菌。16SrRNA系统发育分析表明,14株嗜碱菌划分为3个群:类群I包含11株菌,与假坚强芽胞杆菌Bacillus pseudofirmus的亲缘关系较近,类群II由FJAT-24879组成,与Jeotgalibacillus campisalis的亲缘关系较近;类群III包含2株菌,与Bacillus horikoshii具有较近的亲缘关系。3种芽胞杆菌均在pH8以上生长良好,生理生化结果分析表明嗜碱芽胞杆菌杆菌类群具有丰富的生理生化特征。研究结果表明西藏尼玛县盐碱地含有较多芽胞杆菌种类。 Bacillusspecies in the saline-alkali habitatsat Nima county, Tibet were surveyed for microbial resource collection. The samples obtained were cultured, isolated and preserved. They were identified using 16S rRNA gene sequence, and typical strains selected for bio-characteristic determination. Among the samples, 14 Bacillus strains were alkaliphilic. The 16S rRNA phylogenetie analysis rendered them in 3 clustered groups: Group 1 of 11 strains closely related toBacillus pseudofirmus; Group 2 of one strain; and, Group 3 of 2 strains identified as Bacillus horikoshii. These strains exhibited diverse bio-characteristics. It appeared that a rich resource of alkaliphilic Bacillusexisted in the region.
