摘要
目的建立采用高效液相梯度洗脱法同时测定参甘养心片中延胡索乙素、苦参碱和氧化苦参碱含量的方法。方法使用Welch Ultimate XB-C18(4.6×250 mm,5μm)色谱柱、流动相为乙腈-0.05mol/L磷酸二氢铵溶液梯度洗脱、检测波长为220 nm、柱温30℃、流速为1.0 ml/min。结果延胡索乙素在0.18~2.4μg,苦参碱在0.14~1.92μg,氧化苦参碱在0.11~1.52μg范围内呈良好的线性关系(r〉0.999 8),平均回收率分别为99.07%,99.80%,100.82%,RSD分别为1.79%,1.04%,0.92%。结论该方法能够将延胡索乙素、苦参碱和氧化苦参碱很好的分离,测定结果准确可靠,重现性好,可以作为参甘养心片质量控制的参考依据。
Objective To establish an HPLC method for determinatiofi the three main components of tetrahydropalmatine, matrine and oxymatrine in Shenganyangxin Tablets. Methods The components were performed on a Welch Ultimate XB-C18 column using a mobile phase of acetonitrile - 0.05 mol/L of ammonium dihydrogen phosphate;and the detective wavelength was 220 nm. Results The three components showed good linear relationship (r 〉 0.999 8) in the range of 0.18 - 2.4 μg for tetrahydropalmatine, 0.14 - 1.92 μg for matrine and 0.11 - 1.52 μg of oxymatrine. The average recoveries were 99.07%, 99.80% and 100.82% with RSD 1.79%, 1.04%, 0.92%, respectively. Conclusion This method can separate the tetrahydropalmatine, matrine and oxymatrine perfectly, the results are accurate, reliable and reproducible, it can be used for the quality controlling of Shenganyangxin Tablets.
