摘要
目的:对粉防己多糖(FDT)与汉中防己多糖(HDT)进行比较研究,明确其化学结构与抗氧化活性的异同。方法:采用比色法和气质联用法,对多糖结构进行研究。通过DPPH自由基、羟自由基、超氧阴离子自由基清除实验,探讨其体外抗氧化作用。通过D-半乳糖诱导小鼠衰老模型,以超氧化物歧化酶(SOD)、丙二醛(MDA)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-PX)在血清、肝、脑、心中表达为指标,配合肝脏病理切片探讨体内抗氧化作用。结果:FDT与HDT的组成成分与单糖组成存在差异,体外抗氧化实验FDT强于HDT,体内实验FDT与HDT在血清、脏、脑、心中对SOD、MDA、CAT及GSH-Px具有不同影响(P<0.01,P<0.05)。结论:FDT与HDT存在结构差异,FDT抗氧化作用强于HDT。
Objective: To compare the Stephania tetrandra S. Moore. polysaccharide(FDT) and Aristolochia heterophyllaHemsl. polysaccharide(HDT) and to clarify their differences of structural characteristics and antioxidant effects. Methods: With the colorimetry and GC-MS analysis, the structure of polysaccharides was studied. DPPH radicals, hydroxyl radicals, and superoxide anion radical scavenging experiments were applied to investigate the antioxidant activity in vitro. D-galactose induced mice aging model was used to investigate the expression of SOD, MDA, CAT, GSH-PX in serum, liver, brain, and heart, and the antioxidant effect in vivo with liver biopsy. Results: The ingredient compositions and monosaccharide compositions between FDT and HDT were differences. The antioxidant activities of FDT were stronger than those of HDT in vitro, FDT and HDT showed different effects on partial oxidase in different organs FDT and HDT have different effects on SOD, MDA, CAT and GSH-PX in serum, liver, brain and heart of antioxidant experiments in vivo(P〈0.01, P〈0.05). Conclusion: The structures between FDT and HDT are different and the antioxidant activity of FDT is stronger than HDT.
