摘要
目的:探讨黄芪、三七及其配伍对慢性萎缩性胃炎大鼠Hedgehog(Hh)信号通路的影响。方法:雄性Wistar大鼠随机分为空白组、模型组、黄芪组、三七组、黄芪三七组及Purmorphamine组,运用幽门弹簧法制备慢性萎缩性胃炎模型。应用HE染色评价胃黏膜病理损伤,酶联免疫吸附法检测血清胃蛋白酶原Ⅰ(PGⅠ)、胃蛋白酶元Ⅱ(PGⅡ)及胃泌素(GAS)水平,应用免疫组织化学法、Western bot和RT-PCR分别检测各组大鼠胃组织Hh信号通路的蛋白和基因表达水平。结果:与空白组比较,模型组大鼠血清PGⅠ/PGⅡ和GAS含量,Shh、Ptch、Gli-1基因及蛋白含量,Smo蛋白表达均显著降低(P<0.01,P<0.05);与模型组比较,黄芪三七组PGⅠ/PGⅡ明显升高(P<0.01),黄芪组及Purmorphamine组GAS含量明显升高(P<0.01,P<0.05);黄芪组、三七组、黄芪三七组及Purmorphamine组Shh、Ptch、Gli-1蛋白及基因含量显著升高(P<0.01,P<0.05),黄芪三七组及Purmorphamine组Smo蛋白表达明显升高(P<0.05,P<0.01)。结论:黄芪、三七及其配伍能明显改善慢性萎缩性胃炎大鼠的胃黏膜病变,其治疗机制可能与激活Hh信号通路有关。
Objective: To investigate the effects of Radix Astragali seu Hedysari, Radix Notoginseng and their compatibility on Hedgehog(Hh) signal pathway in chronic atrophic gastritis(CAG) rats. Methods: Male Wistar rats were randomly divided into the control group, model group, Radix Astragali seu Hedysari group, Radix Notoginseng group, Astragali seu Hedysari plus Radix Notoginseng group and Purmorphamine group. CAG model was induced by pylorus spring implanting method. HE staining was used to examine pathological damage of gastric mucosa. The level of PGⅠ, PGⅡ and GAS in serum were detected by ELISA assay. The m RNA and protein expression of indicators in Hh signal pathway were examined by immunological histological chemistry, RT-PCR and Western blot analysis respectively. Results: Compared with the control group, the level of PGⅠ/PGⅡand GAS in serum, m RNA and protein expression of Shh, Ptch and Gli-1, and protein expression of Smo were significantly decreased in model group(P〈0.01, P〈0.05); compared with the model group, the level of PGⅠ/PGⅡ of serum in Astragali seu Hedysari plus Radix Notoginseng group was significantly increased(P〈0.01), and GAS level increased in the Astragali seu Hedysari group and Purmorphamine group(P〈0.01, P〈0.05); compared with the model group, m RNA and protein expression of Shh, Ptch and Gli-1 were significantly increased(P〈0.01, P〈0.05) in four treatment groups, and expression of Smo was significantly increased in the Astragali seu Hedysari plus Radix Notoginseng group and Purmorphamine group(P〈0.05, P〈0.01). Conclusion: Astragali seu Hedysari, Radix Notoginseng and their combination can improve mucosal atrophy in rats with CAG, which may be associated with an activation of Hh signaling pathway.
