摘要
目的 探讨冀产远志与卵叶远志的种间差异,以指导临床合理用药。方法 采用高效液相色谱法测定冀产远志与卵叶远志在不同采集期、不同用药部位中有效活性成分远志山酮Ⅲ与3,6二芥子酰基蔗糖的含量。色谱柱为Sunfire C18(4.6 mm×250 mm,5μm),流动相为甲醇-0.05%磷酸溶液(70∶30),检测波长为230 nm,流速为1.0 m L/min,柱温30℃。结果各组分分离度良好,远志口山酮Ⅲ在0.460034.6003 mg/m L浓度范围内与峰面积线性关系良好(r=0.999);3,6二芥子酰基蔗糖在0.415874.15872 mg/m L浓度范围内与峰面积线性关系良好(r=0.999),平均回收率分别为98.10%(RSD=1.80%)和98.13%(RSD=1.63%),7批样品中远志山酮Ⅲ的含量分别为0.113、0.078、0.121、0.105、0.082、0.054、-0.003 g/g,3,6二芥子酰基蔗糖的含量为0.665、0.392、0.390、0.453、0.394、0.586、0.411 g/g。结论该方法简便、快速,为冀产远志的品种开发提供科学数据。
Objective To discuss the difference between Polygda tenuifolia Willd. and Polygala sibirica L. for guiding clinical rational drug use. Methods Effective active ingredient Polygalaxanthone and 3,6'-Disinapoylsucrose content of different collection period and parts of Polygala tenuifolia Willd and Polygala sibirica L were measured by HPLC method. The determination was performed on Sunfire C18 (4.6 min×250 mm, 5 μm) column with methanol -0.05% phosphate acid (70:30) as mobile phase at detection wavelength of 230 nm, the flow rate was 1.0 mL/min, the column temperature was set at 30℃. Results The components were separated completely. The good linear relationship of Poly- galaxanthone Ⅲ occurred in the range of 0.460 03-4.6003 mg/mL(r = 0.999), the good linear relationship of 3,6'-Disinapoylsucrose in the range of 0.415 87-4.158 72 mg/mL(r = 0.999). The average recoveries were 98.10% (RSD=1.80%) and 98.13% (RSD=l.63%). In the 7 groups, the content of Polygalaxanthone Ⅲ was 0.113, 0.078, 0.121, 0.105, 0.082, 0.054, -0.003 g/g, 3,6'-Disinapoylsucrose was 0.665, 0.392, 0.390, 0.453, 0.394, 0.586, 0.411 g/g. Conclusion The method is simple and quick, which can be used for rational development of Polygala tenuifolia Willd.
