摘要
建立了西兰花籽中萝卜硫苷的高效液相色谱测定方法。色谱条件为:Ecosil C18(200 mm×4.6 mm,5μm)为色谱柱;0.1%甲酸水溶液-甲醇溶液(98:2,V/V)为流动相,等梯度洗脱;检测波长225 nm;流速为1.0 m L/min;柱温30℃。在该色谱条件下萝卜硫苷与其他物质得到了有效的分离,且萝卜硫苷在0.196~7.84μg范围内与峰面积有良好的线性关系,R=0.99997,萝卜硫苷回收率在97.91%~101.02%之间。该方法重复性好、精密度高、回收率较好,可作为西兰花籽提取物中萝卜硫苷的定量测定方法。
A method was developed for the determination of gulcoraphanin in broccoli seed by high performance liquid chro- matograph (HPLC). The optimum condition was: Ecosil C,s column (200 mm×4.6 ram, 5 p,m), mobile phase: 0.1% formic acid-water and methanol solution; wavelength: 225nm; flow rate: 1.0mL/min; column temperature: 30℃. This method showed good linearity for gulcoraphanin (ranging from 0.196 to7.84 p,g), the recovery of gulcoraphanin was from 97.91% to 101.02 %. The results demonstrated that this method was precise, accurate and can be used for the determination of glu- coraphanin in Brassiea oleracea L.var.italic Planch.
