TNF—α、NF-κBp65、PAI-1在妊娠期糖尿病孕妇血清和胎盘组织中的表达及其意义 被引量:14

Expression of TNF-α, NF-κBp65 and PAI-1 in Serum and Placenta Tissues of Pregnant Women with Gestational Diabetes Mellitus and its Clinical Significance

摘要 【目的】探讨肿瘤坏死因子(TNF—α)、人核转录因子p65(NF-κBp65)及纤溶酶原激活物抑制物1(PAI-1)在妊娠期糖尿病(gestational diabetes mellitus,GDM)孕妇血清及胎盘组织中的表达及其临床意义。【方法】选取2014年2月至2015年5月在本院定期产检并且住院分娩的GDM孕妇47例作为观察组,另外选取同期分娩的正常孕妇47例为对照组,比较两组孕妇血清和胎盘组织中TNF-α、NF-κBp65和PAI-1的表达水平,并分析其临床意义。【结果】与对照纽比较,观察纽孕妇血清的TNF-α和PAI-1表达水平明显较高,差异具有统计学意义(P均〈0.001),但两组孕妇血清的NF—KBp65水平比较差异无统计学意义(P〉0.05);观察组孕妇胎盘组织中的TNF-α、NF-κBp65和PAI-1的阳性率显著高于对照组,其差异均有统计学意义(P〈0.05)。【结论】GDM孕妇胎盘组织中TNF—α、NF—κBp65和PAI-1水平明显升高,而血清中的TNF-α和PAI-1也明显较高,推测炎症因子TNF-α、NF-κBp65和PAI-1可能参与了GDM的发生与发展。 [ Objective]To investigate the changes of expression of tumor necrosis factor (TNF-a), human nuclear transcription factor p65 (NF-KBp65), and plasminogen antivator inhibition 1 (PAI-1) in serum and placenta tissues of pregnant women with gestational diabetes mellitus and its clinical significance. [Methods] 47 pregnant women with gesta- tional diabetes mellitus, who received regular antenatal care and delivered in the obstetrical department between February 2014 and May 2015 were selected for the observation group. 47 normal pregnant women who delivered babies in the same period were selected as the control group. The expression levels of TNF-α, NF-KBp65, and PAI-1 in the serum and placental tissues were compared between the two groups. The correlation between expression levels of TNF-α, NF-KBp65, and PAI-1 in the serum was analyzed. [Results]The expression levels of TNF-α and PAI-1 in the serum of the observation group were significantly higher than those in the control group ( P 〈0.001). However, there was no significant difference of NF-KBp65 level in serum between the two groups ( P 〉0.05). The positive detection rates of TNF-α, NF-KBp65, and PAI-1 of the placenta in the observation group were significantly higher than those in the control group ( P %0.05). [Conclusion]TNF-α, NF-KBp65, and PAI-1 in serum and placental tissues may participate in the onset and development of gestational diabetes mellitus.
