芍甘附子汤加味对CIA大鼠关节滑膜组织JAK/STAT通路相关基因表达的影响 被引量:9

摘要 目的:观察芍甘附子汤加味对胶原性关节炎(collagen-induced arthritis,CIA)大鼠关节滑膜组织JAK/STAT通路相关基因SOCS1 mRNA、SOCS3 mRNA以及STAT1 mRNA表达的影响。方法:建立CIA大鼠模型,采用real-time PCR法检测大鼠膝关节滑膜组织SOCS1 mRNA、SOCS3 mRNA以及STAT1 mRNA的表达含量。结果:两组比较Ct值的相对含量,结果显示大鼠滑膜组织正常组,模型组,芍苷附子汤小剂量组、中剂量组、大剂量组,对照组STAT1 mRNA表达相对值分别是1、38.32、12.04、6.15、8.94、8.06;STAT1 mRNA表达相对值分别是1、17.03、5.98、2.50、3.36、4.66;SOCS3 mRNA表达相对值分别是1、15.89、3.58、1.75、2.22、3.12。说明模型组SOCS1 mRNA、SOCS3 mRNA以及STAT1 mRNA表达水平与正常组相比较,均有明显增高(P<0.05),以STAT1 mRNA增高最显著。治疗组和对照组SOCS1 mRNA、SOCS3 mRNA以及STAT1 mRNA表达水平与模型组相比较,均有明显下降(P<0.05)。结论:芍甘附子汤加味能有效调控JAK/STAT信号转导通路STAT1 mRNA、SOCS1 mRNA、SOCS3 mRNA的表达,可能是其实现抗炎效应及CIA大鼠好转的重要分子机制之一。 Objective:To observe adding Shaogan Fuzi Tang to influnence of relative gene expression( SOCS1 mRNA,SOCS3 mRNA and STAT1 mRNA) of the JAK / STAT signaling pathway of the joint synovial tissue in CIA( collagen-induced arthritis) rat.Methods:The rat model of CIA was replicated,and utilize the method of Real-time PCR was used to detect the expression value of rat knee joint synovial tissue SOCS1 mRNA,SOCS3 mRNA and STAT1 mRNA. Results:Compared the Ct value of the two groups,the relative expression value of the joint synovial tissue of the rat of the normal group,modal group,little dose group,medium dose group,large dose group was relatively 1,38. 32,12. 04,6. 15,8. 94,and 8. 06. The relative expression value of STAT1 mRNA was relatively 1,17. 03,5. 98,2. 50,3. 36 and 4. 66. The relative expression value of SOCS3 mRNA was relatively 1,15. 89,3. 58,1. 75,2. 22,and 3. 12. It indicates that the SOCS1 mRNA,SOCS3 mRNA expression value and STAT1 mRNA of the model group were significantly increased( P 〈 0. 05),with the most significant increasing of the STAT1 mRNA. Compared with the model group,the expression value of the SOCS1 mRNA,SOCS3 mRNA and STAT1 mRNA of the treatment group and control group were significantly decreased. Conclusion:Adding Shaogan Fuzi Tang is effective in regulating and controlling the expression of the JAK /STAT signaling pathway of STAT1 mRNA,SOCS1 mRNA and SOCS3 mRNA,which is one of the important molecular mechanism of the antigen effect and the improvement of the CIA rat.
