摘要
目的探讨下丘脑室旁核(PVN)orexin-A对大鼠胃运动的调控作用及其机制。方法选用成年Wistar大鼠48只,随机分为6组,PVN分别微量注射0.5μL生理盐水(NS,A组),0.05、0.50、5.00μg orexin-A(B、C、D组),6.00μg SB334867(E组),0.50μg orexin-A+6.00μg SB334867混合液(F组)。另取大鼠60只,随机分为6组,分别为假手术组(G组)、单纯切断双侧膈下迷走神经组(H组)、假手术+NS 0.5μL组(I组)、假手术+5.0μg orexin-A组(J组)、迷走神经切断+NS 0.5μL组(K组)、迷走神经切断+5.0μg orexin-A组(L组)。将应力传感器缝在胃浆膜层,观察并记录自由活动大鼠胃收缩幅度及频率。结果与A组相比,B、C、D组大鼠给药10-20min胃收缩幅度明显增高,呈明显的剂量依赖关系(F=7.14-13.73,q=2.80-7.39,P〈0.05),且胃收缩频率也呈剂量依赖性增高(F=9.97-13.36,q=2.05-7.30,P〈0.05);与C组相比,F组大鼠的orexin-A促进胃收缩幅度和频率增加作用明显减弱(t=4.52-5.75,P〈0.05);A组和E组大鼠给药后胃收缩幅度和频率比较差异均无显著性(P〉0.05)。与G组相比,H组大鼠胃收缩幅度和频率明显降低(t=7.40-14.35,P〈0.05);与I组相比,J组大鼠胃收缩幅度和频率明显升高(t=3.65-9.48,P〈0.05);K组和L组大鼠的胃收缩幅度和频率比较差异无显著性(P〉0.05)。结论下丘脑PVN微量注射外源性orexin-A可促进大鼠胃运动,该效应可能是通过下丘脑PVN-迷走神经通路来完成的。
Objective To investigate the effect of orexin-A of hypothalamic paraventricular nucleus (PVN) on the regu lation of gastric motility and its mechanism. Methods Forty eight adult Wistar rats were divided into six groups in random. In tra PVN microinjection of 0.5 μL normal saline (group A), 0.05 μg orexin A (group B), 0.50 μg orexin A (group C), 5.00 μg orexin A (group D), 6.00 〉g SB334867 (group E), and 0.50 p.g orexin-A+6.00 gg SB334867 mixture (group F) was done to cre- ate the above six groups. The other 60 rats were recruited and randomized to six groups as: sham operation group (group G), sire pie subdiaphragmatie vagotomy group (group H), sham operation +0.5 μL. NS group (group l), sham operationq-5.0 μg orexin- A group (group J), subdiaphragmatic vagotomy+0.5 〉L NS group (group K), and subdiaphragmatic vagotomy+5.0 #g orexin-A group (group L). A strain gauge was emhedmented in gastric serosa, and the amplitude and frequency of gastric constriction in freely moving rats were observed and recorded. Results Compared with group A, the rats in groups B, C and D group showed increased amplitude of contraction 10-20 min after medication, indicating dose-dependent (F = 7.14 - 13.73, q = 2.80 - 7.39, P 〈 0.05) and the gastric constriction frequency also indicated dose-independent rising up (F=9.97--13.36 ,q=2.05-7.30 ,P〈0.05). Compared with group C, the effect of orexin A on promoting the amplitude and frequency of gastric contraction in group F significantly decreased (t =4.52-5.75, P〈0.05). After medication, the differences in the amplitude and frequency of gastric contraction between groups A and E were not significant (P〉0.05). Compared with group G, the amplitude and frequency of gastric contraction of rats in group H descended dramatically (t = 7.40-14.35, P 〈0.05). Compared with group I, the amplitude and frequency of gastric contraction of rats in group J increased significantly (t = 3.65-9.48,P 〈0.05). The differences in amplitude and frequency of gastric contraction of rats between groups K and L were not significant (P〉0.05). Conclusion Intra-PVN micro-injection of exogenous orexin-A can promote gastric motility in rats, the effect is likely to be completed through the PVN-vagus pathway.
