摘要
目的检测背部创伤模型大鼠不同愈合时间时血管内皮生长因子(VEGF)、血小板源性生长因子(PDGF)及碱性成纤维细胞生长因子(bFGF)的表达,探讨其在创伤愈合过程中的作用。方法 Wistar大鼠30只,手术法切除背部1cm×1cm大小的皮肤全层组织建立背部创伤模型,分别在手术后第3、5、7、10、14天取创面组织检查,应用苏木精-伊红(HE)、Masson、巴氏染色观察创面组织病理改变,免疫组化法检测创面组织VEGF、PDGF和bFGF的表达。结果 HE染色显示,手术后3d时创面炎性细胞浸润明显,毛细血管丰富。Masson染色显示,手术后5d时创面胶原纤维增生明显,缺损创面被填补完整后胶原纤维染色程度减弱。巴氏染色显示,手术后10d时表皮组织完全覆盖创面,14d时创面中心表皮完全再生,可见分化成熟的基底层、棘层等细胞。免疫组化染色显示,VEGF表达在手术后3d时达高峰,之后逐渐下降(F=14.387,P<0.05);PDGF表达在手术后5d时达高峰,之后逐渐下降(F=70.307,P<0.05);bFGF表达在手术后10d时达高峰(F=24.821,P<0.05)。结论大鼠背部创伤模型诱导了愈合过程的发生,VEGF、PDGF和bFGF的差异性表达在创伤愈合过程中发挥重要作用。
Objective To detect the expressions of vascular endothelial growth factor (VEGF), platelet derived growth factor (PDGF) and basic fibroblast growth factor (bFGF), in different healing period of back-wound rat models, and explore their role in the wound healing process. Methods Thirty Wistar rats were entered in this study. A back wound model in rats was cre ated by cutting 1 cm × 1 cm whole thickness of dorsal skin. Tissues from wound surface were obtained--on days 3, 5, 7, 10, and 14 after surgery--for staining of hematoxylin-eosin (HE) staining, Masson's Trichrome and Papanieolaou (PAP) to observe the pathology of the wound surface, and using immunohistochemistry method to detect the expressions of VEGF, PDGF and bFGF. Results HE staining indicated, 3 days after surgery, obvious inflammatory cell infiltration on wound surface with abundant capillaries. The Masson's Trichrome staining suggested that, 5 days after surgery, the collagen fibers increased significantly, and after the wound was completely healed, the staining of the fibers weakened. The PAP staining revealed that, 10 days after surgery, the wound surface was completely covered by epidermal tissue, and on day 14, regeneration of epidermis was completed on the center of the wound, and full-grown basilar cells, prickle cells and others could be seen. Immunohistochemistry staining demonstrated that the VEGF reached its peak 3 days after surgery, and then gradually declined (F= 14.387 ,P〈0.05) ; PDGF reached its peak 5 days after surgery, and then gradually declined (F=70.307 ,P〈0.05) ; hFGF reached its peak 10 days after surgery (F = 24.821, P〈 0.05). Conclusion The model of back wound in rats induces the occurrence of healing process, and the differential expressions of VEGF,PDGF and bFGF play an important role in the process of wound healing.
