摘要
以君子兰新鲜花粉为材料,采用TTC、I-KI、醋酸洋红和丙酸洋红染色法在相同条件下观测同一批新鲜花粉的生活力,结果表明:采用TTC染色时,全部花粉均未有染色反应(不含脱氢酶);采用I-KI染色时,生活力为85.4%;采用醋酸洋红染色时,生活力为88.1%;采用丙酸洋红染色时,生活力为81.4%。醋酸洋红染色效果最佳,但染色液配制操作复杂;I-KI染色效果稍差,易于操作。
The same batch fresh pollen vitality was tested with TTC, I - KI, aceto - carmine and propionic acid magenta staining method in the same condition by choosing fresh pollen of Clivia miniata as material. The result showed that the whole pollen didn't present staining reaction when it was taken TTC. The vitality was 85.4% , 88.1% , 81.4% by taking I - KI, aceto - carmine and propionic acid magenta staining method, respectively. The staining effect of aceto - carmine was the best while the staining solution preparation operation was complicated. However, the staining effect of I - KI was a little worse while it was easy to operate.
