摘要
探讨绿色木霉固态发酵产生纤维素酶的诱导促进条件。通过初始培养基、表面活性剂、诱导物、缓冲pH体系等对绿色木霉固态发酵产酶的诱导情况进行分析,获得产酶时间曲线和最大产酶量。不同条件下绿色木霉产纤维素酶活力存在显著差异(P<0.05),麸皮∶秸秆粉=1.5∶1(质量比),培养基固液比1∶2.5(g/m L),硫酸铵按培养基干基的2%加入,初始pH=6为最佳条件,采用缓冲pH体系对产酶有促进作用,添加表面活性剂和适当种类的诱导物可以提高产酶量,而过高浓度的诱导物则降低产酶量。该研究为绿色木霉对秸秆的转化应用提供了理论依据。
To explore promoting conditions of solid fermentation degrading corn straw by Trichoderma viride.Using Trichoderma viride as experimental materials,the conditions for cellulose production were studied by solid fermentation. The influences of the culture conditions,including medium,a surfactant,inducers,pH buffering system,enzyme-time curve and maximum enzyme production was obtained. The result showed that there was significant difference in cellulase activity produced by Trichoderma viride under different conditions(P0.05).It was found that optional conditions for fermentation of corn straw were based on culture at wheat bran and corn straw in the ratio of 1.5 ∶ 1(mass ratio),under 6.0 pH,with the medium liquid of 1 ∶ 2.5(g/m L),according to the medium dry ammonium sulfate 2 % was added. Using the pH buffer system for enzyme production advantageously,a surfactant is added and appropriate type of inducer enzyme production could be improved,and the high concentration of inducer may decrease the amount of enzyme production. The research provided certain theoretical basis for the transformations and the application of Trichoderma viride on straw.
