摘要
分别用2 Gy和4 Gy^(60)Coγ射线照射人肝细胞HepG2,利用real-time PCR方法检测PIF1基因mRNA表达量的变化,以探讨PIF1基因是否参与电离辐射致DNA损伤应答。结果表明,不同剂量的γ射线照射HepG2细胞后,PIF1 mRNA表现出一致的变化规律:即照后先轻微降低随后一直高表达,高表达至少持续到照射后12 h。提示电离辐射能够诱导PIF1表达,表达水平与剂量、时间相关。
Hep G2 cells were irradiated by 2 Gy and 4 Gy^60Coγ ray,the mRNA was extracted and real-time PCR was used to detect the changes of PIF1 mRNA,to explore whether PIF1 involved in DNA damaged respond after irradiation. The results showed that PIF1 gene expression level in Hep G2 decreased slightly at first and then increased after different doses of ^60Coγ ray,and the high expression continued about 12 h. PIF1 gene expression can be induced by ionizing radiation indicating its roles in DNA damage response,and the expression level is related to radiation doses and time.
