C反应蛋白和白细胞介素6在自然衰老大鼠血管中的表达研究

C-reactive protein and interleukin-6 levels in normal aging rats aorta

摘要 目的:研究炎性因子C反应蛋白(CRP)和白细胞介素(IL)-6在不同年龄组大鼠主动脉组织中的表达差异,及其可能在血管衰老机制中的作用。方法:清洁级雄性SD大鼠24只,其中12只青年大鼠(青年组,2月龄)和12只老年大鼠(老年组,20月龄),经适应性喂养1周后麻醉采静脉血待测,处死后取胸主动脉组织,石蜡包埋切片HE染色后光镜下观察血管组织形态改变,Western blot检测主动脉p16和p21蛋白水平,化学发光酶免疫分析法和酶联免疫吸附试验(ELISA)分别测定大鼠血清CRP和IL-6的含量。结果:老年组大鼠主动脉组织HE染色血管中膜层增厚、血管平滑肌细胞排列紊乱,Western blot检测p16和p21蛋白表达阳性;青年组大鼠主动脉组织HE染色血管中膜层正常、血管平滑肌细胞排列整齐,p16和p21蛋白表达阴性。与青年组大鼠相比,老年组血清CRP和IL-6含量均明显升高(均P<0.05)。结论:炎症可能是导致血管衰老的原因之一。 Objective:To explore the expression of C-reactive protein(CRP)and interleukin-6(IL-6)in aortic of aging rats and the possible effect on vascular aging.Method:Twenty-four male SD rats were composed of 12 young rats(2months old)and 12 elderly rats(20months old).After 1-week feeding for adaptability,blood was collected from vein under anesthesia and the aorta was took out after the rats were sacrificed.Light microscope was used to observe structural changes of aorta,the protein level of aorta p16 and p21were detected by Western blot,chemiluminescence enzyme immunoassay was used to examine serum CRP,and the level of serum IL-6was detected by ELISA.Result:HE staining in aorta tissue showed thickened tunica media and disordered vascular smooth muscle cells in elderly group,however,vessel wall of aorta in young group rats were normal,which with vascular smooth muscle cells regularly.The protein level of p16 and p21in aorta was significantly increased in elderly group.Compared with young group,the level of serum CRP and IL-6were higher in elderly group(both P〈0.05).Conclusion:Higher expression of CRP and IL-6in aging rats aortic is closely correlated with inflammation may be play an important role on vascular aging.
