腰痛颗粒剂对自体髓核移植大鼠痛阈及PGE2、6-Keto-PGF1a的影响

The Influence of Lambago Granules on Pain Threshold and PGE2 and 6-Keto-PGF1a of Rats with Transplantation of Autologous Nucleus Pulposus

目的:探讨腰痛颗粒剂对自体髓核移植致腰神经根性疼痛模型大鼠痛阈及PGE2、6-Keto-PGF1a的影响。方法:将32只SD大鼠随机分成腰痛颗粒剂组、腰痛宁组、假手术组和空白对照组,每组各8只。建立大鼠自体髓核移植致腰神经根性疼痛模型,腰痛颗粒剂组、腰痛宁组分别给予腰痛宁灌胃;假手术组给予凉开水灌胃;空白对照组给予正常饮食。分别于造模前、造模后7 d、造模后21 d观察大鼠行为学及痛阈的变化,造模后21 d后处死大鼠心脏取血,采用酶联免疫吸附法测量血清中PGE2、6-Keto-PGF1a含量。结果:给药后腰痛颗粒剂组、腰痛宁组大鼠PWT、PWL均较给药前明显降低,且腰痛颗粒剂组优于腰痛宁组,差异均有统计学意义(P<0.05);给药后腰痛颗粒剂组PGE2、6-Keto-PGF1a含量明显低于腰痛宁组,差异具有统计学意义(P<0.05)。结论:腰痛颗粒剂可提高自体髓核移植大鼠的痛阈。

Objective：To explore the influence of Lambago Granules on pain threshold and PGE2 and 6-Keto-PGFla of model rats with transplantation of autologous nucleus pulposus leading to lumbar nerve root pain. Methods：Thirty-two SD rats were randomly divided into the Lambago Granules group, the Yaotongning （YTN） group, the sham-surgery group and the control group, with 8 rats in each group. The Lambago Granules group and the YTN group were given Lambago Granules and YTN respectively through administration by gavage ;The sham-surgery group was given cold boiled water while the control group was given normal diets. The rats＇ behaviours and pain threshold changes before establishing the model,7 days and 21 days later after establishing the model were observed respectively. Twenty-one days later after establishing the model, they were killed and their heart blood was taken. Then contents of PGE2,6-Keto-PGF1 a in serum were measured by using enzyme-linked immunosorbent assay. Results：After being given medicine, PWT and PWL of the Lambago Granules group and the YTN group were obviously decreased than before the treat- ment;and the Lambago Granules group was better than the YTN group;And the differences between them had statistical signifi- cance （ P 〈 0.05 ）. After being given medicine, PGE2,6-Keto-PGF1 a contents of the Lambago Granules group were obviously low- er than those of the YTN group ; And the differences between them had statistical significance （ P 〈 0.05 ）. Conclusion ： Lambago Granules can improve the pain threshold of rats with transplantation of autologous nucleus pulposus
