摘要
目的:观察苗药通窍化栓汤对MCAO大鼠急性期脑梗死区VEGF表达的影响。方法:将60只SD大鼠随机分为苗药组和对照组,每组30只。均采用线栓法将SD大鼠右侧大脑中动脉皮质支闭塞(middle cerebral artery occlusion,MCAO),在MCAO造模成功再灌注后6 h,苗药组给予通窍化栓汤中药液,每次5 m L,每日1次,对照组给予复方丹参注射液,剂量0.01 m L·g-1,每日1次。于给药后第2周处死取脑,用Western蛋白印迹检测两组大鼠脑梗死区血管内皮生长因子(vascular endothelial growth factor,VEGF)表达的变化,并观察两组大鼠给药前后神经功能缺损的变化。结果:造模成功后,给药前两组神经功能缺失评分无显著性差异(P>0.05);给药后两组神经功能缺失评分较前均有改善,且苗药组优于对照组(P<0.05);Western蛋白印迹检测提示两组均促进VEGF在梗死区的表达,但苗药组明显优于对照组(P<0.05)。结论:通窍化栓汤对MCAO大鼠梗死区VEGF的表达具有明显促进作用。
Objective:To observe the influence of Tongqiao Huashuan Decoction of Miao medicine on VEGF expression of cerebral infarction area in acute stage of MCAO rats. Methods:Sixty SD rats were randomly divided into the Miao medicine group and the control group, with 30 in each group. All the rats in the two groups were made into the model of middle cerebral artery occlusion (MCAO) by using suture method, then they were infused for 6 hours. The rats in the Miao medicine group were treated with Tongqiao Huashuan Decoction (TQHQD) ,5 mL each time and once a day,while the ones in the control group were given Com- pound Danshen Injection ,0. 01 mL· g-1 and once a day. They were killed and their brains were taken out 2 weeks later after they were given medicine. Then the two groups' vascular endothelial growth factor (VEGF) expression changes were detected by using Western blot technic, and their neural function defect changes before and after the treatment were also observed. Results:After having establishing the model successfully,before they were given medicine,the two groups' neural function defect scores had no obvious difference (P 〉 0. 05 ). After they were given medicine ,the two groups' neural function defect scores were both improved than before the treatment ; And the Miao medicine group was better than the control group ( P 〈 0.05 ). Western blot technic showed that the two groups promoted VEGF expression of cerebral infarction area, but the Miao medicine group was much better than the control group ( P 〈 0.05 ). Conclusion : TQHQD can obviously promote VEGF expression of cerebral infarction area ofMCAO rat.
