风湿骨痹口服液治疗兔膝骨性关节炎实验研究

The Experimental Research into Rabbit's Knee Arthritis Treated with Oral Liquid of Rheumatoid Arthralgia

目的:观察风湿骨痹口服液对实验性家兔膝骨性关节炎的治疗作用及其作用机制。方法:将30只新西兰大耳兔随机分为正常组、模型对照组及药物组,每组各10只。采用Hulth法复制膝骨性关节炎模型,造模1周后,正常组和模型组用生理盐水,药物组用风湿骨痹口服液灌胃连续给药6周,经大体观察后,测定各组血清中的一氧化氮(nitric oxide,NO)、超氧化物歧化酶(Superoxide Dismutase,SOD)和丙二醛(malonaldehyde,MDA)的含量;并测定关节液中肿瘤坏死因子-α(tumor necrosis factor,TNF-α)、白介素-1(interleukin-1,IL-1)、基质金属蛋白酶-1(Matrix metalloproteinase,MMP-1)的含量。结果:与模型组比较,药物组能够降低血清中NO和MDA含量,并提高SOD活性;关节液中IL-1、TNF-α和MMP-1含量明显降低,差异具有统计学意义(P<0.05)。结论:风湿骨痹口服液对实验性家兔膝骨性关节炎具有治疗作用,其作用机制可能是通过有效抑制细胞因子、基质金属蛋白酶、氧自由基的生物效应来延缓软骨的退变。

Objective ：To observe the clinical curative effect and its working mechanism of Oral Liquid of Rheumatoid Arthralgia on rabbits＇ knee arthritis. Methods： Thirty New Zealand big-eared rabbits were randomly divided into the normal group, the model group and the medicine group, with 10 cases in each group. The knee arthritis model was copied by using Hulth method. One week later, the ones in the normal group and the model group were given normal saline, while the ones in the medicine group were given Oral Liquid of Rheumatoid Arthralgia respectively by perfusion for 6 weeks continuously. After a general observation,each group＇ s nitric oxide（NO） , Superoxide Dismutase（SOD） and malonaldehyde（MDA） contents as well as contents oftumor necrosis factor （TNF-ot） , interleukin-t, （ IL-1 ） and Matrix metalloproteinase （ MMP-1 ） in joint fluid were measured. Results ： Compared with the model group, the medicine group could reduce contents of NO and MDA in serum,but improve SOD activity;The contents of IL-1, TNF-a,MMP-3 obviously decreased;And all the differences between them had statistical significance（ P 〈 0.05 ）. Conclusion：O- ral Liquid of Rheumatoid Arthralgia has a remarkable clinical curative effect on knee arthritis of experimental rabbits, and its working mechanism may have something to do with delaying the degeneration of cartilage by effectively inhibiting cell factor, ma- trix metalloproteinases and biological effects of oxygen free radicals.
