DEB-2产抗真菌活性物质发酵条件及有机试剂对其活性的影响

摘要 为探索DEB-2菌株生产生物农药的工业发酵条件,采用活性检测方法,对DEB-2菌株的发酵培养基及其制备工艺、发酵罐的最适宜发酵时间,发酵液剂型的基础条件如干燥方法、干燥时间和有机试剂对抗菌物质的影响等进行研究。结果表明:最佳培养基为PD培养基,最佳铃薯汁液量为30%。在培养基制备工艺的筛选中,马铃薯打浆法与马铃薯煮汁法的效果相当。在100 L发酵罐中PD培养基的装液量为70%、发酵温度35℃、发酵时间为40.2 h时发酵液的抗菌活性达到最高,抑制率达100%。滤纸过滤和8000 r/min离心10 min处理发酵液对其活性物质几乎没有影响;冻干处理的发酵液中活性物质较烘干的损失小,其抗菌活性损失为4.77%。用50%浓度的甲醇、乙醇、丙酮处理DEB-2菌的发酵液,甲醇对其活性影响最小,其上清液的抗菌活性与发酵原液相当。 The effect of fermentation media,preparation technology,suitable fermentation hour,basic conditions of fermentation broth form such as drying method,drying hour and organic reagents on antifungal activity substance produced from DEB strain by the activity detection method to discuss the biopesticide industrial fermentation conditions of DEB strain. Results: The optimum medium is PD medium and the optimum potato juice amount is 30 %. The effect of potato mashing method is similar to potato boiled juice method. The antifungal activity of DEB strain under the fermentation conditions of 70 % loading liquid PD medium in 100 L fermentation cylinder,35 ℃ and 40. 2h is up to the maximum and its inhibition rate reaches 100 %. The fermentation broth treated by the filter paper and centrifugal method with 8000 r / min for 10 min had little or no effect on activity substance of DEB strain. The activity substance loss of the fermentation broth treated by frozen dried method is lower than the drying method and the loss of antifungal activity is 4. 77 %. Methanol has the minimum effect on antifungal activity of DEB strain fermentation broth among methanol,ethanol and acetone treatment and the antifungal activity of the supernatant liquor is similar to the original fermentation broth.
