脑缺血大鼠海马和心肌组织α-actinin蛋白表达及“脑心同治”方的干预作用 被引量:6

摘要 目的:探讨α-辅肌动蛋白(α-actinin)在脑缺血大鼠海马及心肌组织中表达的变化情况,并观察"脑心同治"方对其表达的影响。方法:采用大脑中动脉堵塞(MCAO)法制作大鼠脑缺血模型,随机分为假手术组、模型组、丹红组、金纳多组。分别对各组进行大鼠神经功能评分,脑梗死体积、脑水含量及血脑屏障通透性检测,并采用Western blot方法检测海马及心肌组织α-actinin蛋白表达。结果:脑缺血大鼠神经功能缺失,脑梗死率、脑水含量增加(P<0.01),血脑屏障通透性增大(P<0.01),海马及心肌组织α-actinin含量降低(P<0.01);药物干预后,神经功能评分降低,脑梗死率、脑水含量及血脑屏障通透性有明显改善(P<0.01),并对海马组织α-actinin蛋白含量有显著调整作用(P<0.05,P<0.01),另外丹红注射液对脑缺血状态下心肌组织中的α-actinin蛋白含量改变亦有纠正作用(P<0.01)。结论:α-actinin蛋白表达的降低可能参与了脑缺血大鼠血脑屏障通透性的改变,"脑心同治"方可以上调MCAO大鼠脑及心肌组织α-actinin蛋白的表达,推测α-actinin蛋白很可能是心脑血管疾病的病理基础及脑心同治应用研究方面的潜在靶点。 Objective:To study the α-actinin expression of hippocampus and myocardial tissue in ischemic stroke rats and the influence of ‘treatment both brain and heart' method.Methods:Middle cerebral artery occlusion model in rats were established by suture-occluded method.Rats were randomly divided into sham, model, Danhong and Ginaton Injection groups, the neurologic deficit score, volume of cerebral infarction, cerebral water content, blood brain barrier(BBB) permeability were measured, the α-actinin protein expression of hippocampus and myocardial tissues were detected by Western blot.Results:The neurologic deficit score, volume of cerebral infarction, cerebral water content and BBB permeability were increased and α-actinin protein expression of hippocampus and myocardial tissue were decreased in ischemic stroke rats(P<0.01).After drug intervention, the neurologic deficit score, volume of cerebral infarction, cerebral water content, BBB permeability and α-actinin protein expression in hippocampus tissue were significantly improved(P<0.05, P<0.01), and Danhong Injection also had improvement to α-actinin protein expression in myocardial tissue(P<0.01).Conclusion:In cerebral ischemic rat, the low protein expression of α-actinin might be involved in BBB permeability change.‘Treatment both brain and heart' method could raise α-actinin expression of hippocampus and myocardial tissue and speculate that α-actinin protein is likely to be the basis of cardiovascular and cerebrovascular diseases and it may be a potential target to ‘treatment both brain and heart' method.
