补肾益精方对RCS大鼠骨髓来源干细胞动员作用的研究 被引量:5

摘要 目的:研究补肾益精方对RCS大鼠骨髓来源干细胞的动员作用。方法:采用先天性视网膜色素变性RCS大鼠作为实验模型,将RCS大鼠随机分为蒸馏水组和和补肾益精方(BSYJ)组;分别予蒸馏水及BSYJ水煎剂灌胃(8.8g/kg),每日1次,以SD大鼠作为正常组。分别于干预7、14、28d进行指标观察,流式细胞仪检测外周血中骨髓来源干细胞(BMSCs)数量;ELISA检测外周血粒细胞集落刺激因子(G-CSF)、基质细胞衍生因子-1(SDF-1)及CXC趋化因子受体4(CXCR-4)含量变化。结果:流式细胞仪检测结果显示,与蒸馏水组相比较,治疗28d时,BSYJ方组外周血BMSCs数量明显增加(P<0.05)。ELISA检测显示,与蒸馏水组相比较,BSYJ方治疗28d后,外周血G-CSF明显增加(P<0.01);BSYJ方治疗14、28d后,CXCR-4含量明显增加(P<0.01)。结论:BSYJ方对RCS大鼠自体BMSCs具有一定的动员作用,这可能与其增加外周血G-CSF及CXCR-4含量有关。 Objective:To investigate the effects of Bushen Yijing(BSYJ) Formula on mobilization of bone marrowderived stem cells in Royal college of surgeons(RCS) rats.Methods:The RCS rats were used as experimental animal model of retinal degeneration.The RCS rats were randomly divided into two groups:the distilled water group and the BSYJ group, while SD rats were used as normal control group.The rats of BSYJ group were given the treatment of BSYJ Decoction(8.8g/kg) by gavage, and the rats of distilled water group were given with distilled water.On the 7th, 14 th and 28 th day after the treatment, the numbers of bone marrow-derived stem cells(BMSCs) in peripheral blood were determined by Flow Cytometry, and the levels of granulocyte colony stimulating factor(G-CSF), stromal cell derived factor-1(SDF-1) and CXC chemokine receptor-4(CXCR-4) in peripheral blood were detected by ELISA assay.Results:The results of Flow Cytometry showed that the treatment of BSYJ Decoction significantly increased the numbers of BMSCs in the peripheral blood on the 28 th day when compared with the distilled water group(P<0.05).The results of ELISA showed that the treatment of BSYJ Decoction significantly increased the levels of G-CSF in the peripheral blood on the 28 th day when compared with the distilled water group(P<0.01), and remarkably enhanced the levels of CXCR-4 on the 4th and 28 th day(P<0.01).Conclusion:BSYJ Formula has a mobilization effect on BMSCs in RCS rats, which is associated with the increased levels of G-CSF and CXCR-4 in peripheral blood.
