当归-红花配伍化学成分UHPLC-Q-TOF-MS分析及配伍协同增效作用研究 被引量:15

Study on chemical component and synergetic effect of concerted application of Chinese angelica and safflower by using UHPLC-Q-TOF-MS

摘要 目的:探讨当归-红花配伍化学成分变化规律及其配伍协同增效作用。方法:采用UHPLC-Q-TOF-MS分析方法,质谱数据采用Marker Lynx 4.1软件进行峰提取、峰对齐及归一化等处理,利用主成分分析法(PCA)和正交偏最小二乘-判别分析法(OPLS-DA)对当归-红花配伍合煎液及单煎合并液的化学成分变化进行比较分析;建立小鼠血虚模型,对单味药材当归、红花及当归-红花配伍的补血作用进行比较研究;建立大鼠急性血瘀模型,对单味药材当归、红花及当归-红花配伍的活血化瘀作用进行比较研究。结果:UHPLC-Q-TOF-MS分析结果显示,当归-红花配伍合煎液与单煎合并液差异明显,当归-红花配伍有利于有效成分的溶出。整体动物评价实验结果显示,当归-红花配伍补血活血作用较优。结论:阐明当归-红花配伍的物质基础,揭示中药配伍协同增效的合理性。 Objective: Study on UHPLC-Q-TOF-MS analysis of chemical constituents and synergistic effects of Danggui-Honghua decoction. Methods: Using UHPLC-Q-TOF-MS analysis method, Marker Lynx 4.1 software processing analysis, principal component analysis(PCA), and orthogonal partial least squares discriminant analysisn(OPLS-DA), chemical constituents changes were compared and analyzed between Danggui-Honghua decoction, Danggui decoction and Honghua decoction. To build rats' blood-deficient mode, the effects of enriching the blood of Danggui-Honghua decoction, Danggui decoction and Honghua decoction were compared. To build rats' acute blood stasis mode, the effects of promoting blood circulation of Danggui-Honghua decoction, Danggui decoction and Honghua decoction were compared. Results: UHPLC-QTOF-MS analysis results showed that the contents of chemical constituents in Danggui-Honghua decoction were higher than the Danggui decoction and Honghua decoction. Whole animal experiment results showed that the effects of enriching the blood and promoting blood circulation of Danggui-Honghua decoction were better. Conclusion: The study was to clarify the material basis of Danggui-Honghua decoction, reveal the rationality of compatibility.
