温通活血乳膏对糖尿病周围神经病变大鼠氧化应激相关因子的影响 被引量:4

Effects of Wentong Huoxue Cream on factors related to oxidative stress in diabetic peripheral neuropathy rats

摘要 目的:建立大鼠糖尿病周围神经痛性病变(DPN)模型,并观察温通活血乳膏对模型动物行为学、坐骨神经传导速度及Bax、Bcl-2表达的影响。方法:用链脲佐菌素(STZ)造Wistar大鼠DPN模型,并随机分为6组:模型组,二甲双胍组(0.18g/kg),辣椒碱乳膏组(0.2g/只),二甲双胍阳性药对照组(辣椒碱乳膏0.2g/只+二甲双胍0.18g/kg),温通活血乳膏低剂量组(0.4g/只+二甲双胍0.18g/kg)、温通活血乳膏高剂量组(0.8g/只+二甲双胍0.18g/kg),各组分别予不同药物给药,连续用药4周后测定各组大鼠的行为学指标、坐骨神经传导速度,同时采用免疫组化法测定各组大鼠的Bax、Bcl-2表达。结果:与模型组比较,温通活血乳膏低、高剂量组大鼠热缩足潜伏期、冷缩足持续时间、机械痛缩足持续时间均延长(P<0.01,P<0.05),缩足反应百分率降低(P<0.01,P<0.05)。与二甲双胍组比较,温通活血乳膏低、高剂量组的冷缩足持续时间延长(P<0.01),温通活血乳膏高剂量组机械痛缩足持续时间延长(P<0.05)。与溶媒对照组比较,模型组大鼠的Bax表达增强(P<0.01),Bcl-2表达减弱和Bcl-2/Bax比值明显降低(P<0.01)。与模型组比较,各药物组坐骨神经细胞Bax表达均降低(P<0.05),Bcl-2表达均增加(P<0.01,P<0.05),以温通活血乳膏高剂量组最明显。结论:温通活血乳膏通过上调Bcl-2表达、下调Bax表达,抑制DPN大鼠坐骨神经细胞凋亡,起到改善神经传导及感觉障碍的作用。 Objective: To establish the model of diabetic peripheral neuropathy in rats, and observe the effects of Wentong Huoxue Cream on model animal behavior, sciatic nerve conduction velocity, Bax and Bcl-2 expression. Methods: With chain urea with cephalosporins(STZ) to build the Wistar rat model of diabetic peripheral neuropathy, and randomly divided into 6 groups, model group, metformin group(0.18g/kg), capsaicin cream group(0.2g/only), metformin, positive medicine control group(0.2g/capsaicin cream+metformin only 0.18g/kg), Wentong Huoxue Cream low dose group(0.4g/metformin+0.18g/kg), Wentong Huoxue Cream high dose group(0.8g/metformin+0.18g/kg), each group separately to different drug dosing for 4 weeks, continuous administration behavior index and sciatic nerve conduction velocity, while Bax and Bcl-2 expression evaluated by immunohistochemical method and Bax. Results: Compared with the model group, foot latency, thermal contraction duration, foot mechanical paw withdrawal duration in Wentong Huoxue Cream low and high dose group were prolonged(P<0.01, P<0.05), and paw withdrawal was decreased(P<0.01, P<0.05). Compared with metformin group, the cold foot duration in Wentong Huoxue Cream low and high dose group was prolonged(P<0.01), mechanical paw withdrawal duration in Wentong Huoxue Cream high dose group was prolonged(P<0.05). Compared with the solvent control group, the expression of Bax in the model group was increased(P<0.01), and the expression of Bcl-2 and Bcl-2/Bax were significantly decreased(P<0.01). Compared with the model group, the expression of Bax in the sciatic nerve cells of each drug group was decreased(P<0.05), and the expression of Bcl-2 was increased(P<0.01, P<0.05). The effects of Wentong Huoxue Cream high dose group was better. Conclusion: Wentong Huoxue Cream can improve nerve conduction, and the function of sensory disturbance by up-regulating Bcl-2 and down-regulating Bax to reduce DPN rats sciatic nerve cell apoptosis.
